早胜牛及其杂交群体SCD1基因外显子5多态性研究 被引量:1

摘要 以5个早胜牛类群(庆阳类群、平凉类群、南德温与庆阳类群杂种、西门塔尔与平凉类群杂种、秦川牛与平凉类群杂种)为研究对象,采用PCR-SSCP及DNA测序方法检测SCD1基因外显子5的多态性。结果表明,早胜牛及其杂交群体SCD1基因外显子5c.878C>T位点发生了C/T的突变,检测到2种基因型(CC和CT)和2个等位基因(C和T),且5个类群均处于中度多态水平。相关性分析表明,早胜牛平凉类群及其余西门塔尔的杂交类群胴体品质和肉质性状在该基因c.878C>T位点的CT和CC基因型之间差异不显著(P>0.05)。因此,SCD1基因外显子5突变位点可作为早胜牛及其杂交群体选育、加强胴体和肉质性状遗传效应的分子标记。 In this study,five Zaosheng cattle groups(Qingyang group,QZ;QZ and its hybrid with South Devon group,QZxN;Pingliang group,PZ;PZ and its hybrid with Qinchuan cattle group,PZxQ;PZ and its hybrid with Simmental group,PZxS) were investigated using PCR-SSCP and DNA sequence analysis to detect polymorphism of SCDl gene exon5 and analyze the correlation between genetic diversity and variation sites,carcass quality and meat quality traits.The results showed that for the SCDl gene in five cattle groups,two genotypes(CC and CT) representing two specific alleles(C and T) in exon5 were detected,with sequence analysis revealing one nucleotide substitution,c.878C〉T.5 groups were in moderate polymorphisms levels.Correlation suggested that Zaosheng cattle Pingliang groups and the rest of the cattle Simmental hybrid taxa had no significant difference between CT and CC genotype in the gene c.878C 〉T site(P〉0.05).Therefore,this mutation of SCDl gene exon5 could be considered as a locus to breed and strengthen carcass and meat quality.
