四君子汤及其拆方对胃癌SGC-7901侧群细胞增殖、侵袭能力的影响

Effects of Sijunzitang and its demolition party on the vitro proliferation and invasiveness of the side population of gastric cancer cell line SGC-7901

目的:探讨含四君子汤及其拆方的药物血清对胃癌细胞株SGC-7901侧群细胞(side population,SP)增殖、侵袭能力的影响。方法:制作含四君子汤及其拆方的兔药物血清;流式细胞仪分选体外培养的胃癌SGC7901细胞株中的SP;平板克隆实验观察含四君子汤及其拆方的药物血清干预后SP细胞与非侧群细胞(non side population,NSP)体外增殖能力的变化;Transwell侵袭实验观察含四君子汤及其拆方的药物血清干预后SP细胞与NSP细胞侵袭能力的变化;比较药物血清干预后SP与NSP细胞的不同。比较四君子汤与其各拆方对SP与NSP细胞干预能力的强弱。结果:胃癌SGC-7901细胞株中分选出SP,比例为(2.68±0.58)%;含四君子汤及其拆方药物血清干预后,SGC-7901 SP与NSP细胞体外增殖、侵袭能力均受到抑制(P<0.05);相同干预条件对SGC-7901 SP与NSP细胞体外增殖、侵袭能力的影响有差异(P<0.05)。四君子汤的抑制作用强于各拆方。结论:胃癌SGC-7901细胞株中存在且能成功分选出SP;含四君子汤及其拆方的药物血清能够抑制胃癌SGC-7901细胞株SP的体外增殖、侵袭能力;药物血清干预后,SP细胞的增殖、侵袭能力仍强于NSPo四君子汤对SP、NSP细胞的抑制作用均强于各拆方。

AIM：To investigate the effects of Sijunzitang and its demolition party drug serum on the proliferation and invasiveness of the side population（SP） of gastric cancer cell line SGC-7901.METHODS：Sijunzitang and its demolition party rabbit serum drug were made;the gastric cancer cell line SGC-7901 SP cells were sorted out by flow cytometry technique;the effects of Sijunzitang and its demolition party drug serum on the vitro proliferation of the gastric cancer cell line SGC-7901 SP cells and non side popution（NSP） cells by tablet cloning experiment were observed;the effects of Sijunzitang and its demolition party drug serum on the invasiveness of the gastric cancer cell line SGC-7901 SP and NSP cells by Transwellexperiment were observed;the difference of SP and NSP cells were compared after drug intervention.The interference abilities of Sijunzitang and its demolition party on SP and NSP cells were compared.RESULTS：The percentage of side group in gastric cancer cell line SGC-7901 was（2.68±0.58）%.The vitro proliferation and invasiveness abilities of Gastric cancer cell line SGC-7901 SP cells and NSP cells were inhibited in Sijunzitang and its demolition party drug serum（P〈0.05）;there was difference between SP and NSP under the same condition（P0.05）;Sijunzitang inhibitory effect was stronger than its demolition party（P0.05）.CONCLUSION：Gastric cancer SGC-7901 cell lines can be successfully sorted out of SP cells;the vitro proliferation and invasiveness of the gastric cancer cell line SGC-7901 SP cells can be suppressed with Sijunzitang and its demolition party drug serum;after the intervention of drug serum,SP cells proliferation and invasiveness ability is stronger than the NSP cells.Sijunzitang inhibitory effect is stronger than its demolition party on SP and NSP cells.
