摘要
目的测定虎杖苷、白藜芦醇、大黄素的平衡溶解度及表观油水分配系数(lg Papp)。方法采用高效液相色谱法测定虎杖苷、白藜芦醇、大黄素在不同p H缓冲溶液中的平衡溶解度;采用摇瓶法结合高效液相色谱法测定其在正辛醇-水及缓冲盐溶液中的表观油水分配系数。结果虎杖苷、白藜芦醇均在p H为6.8的磷酸盐缓冲液中溶解度最大,大黄素在p H为6.5的磷酸盐缓冲液中溶解度最大;虎杖苷、白藜芦醇、大黄素在正辛醇饱和的水中lg Papp值分别为0.98、2.69、0.99,在不同p H的磷酸盐缓冲液中,虎杖苷、白藜芦醇与大黄素的油水分配系数范围分别为0.84-1.01、2.34-2.89、0.97-1.27。结论在胃肠道生理条件下,虎杖苷与大黄素的吸收较好,白黎芦醇的吸收较差;从溶解性能看,大黄素的溶解性能较差,可改善其溶解性能进一步提高其生物利用度。
AIM To determine the equilibrium solubility of polydatin,resveratrol and emodin in different solvents and their n-octanol / water partition coefficients. METHODS The equilibrium solubilities of polydatin,resveratrol and emodin in solutions were determined through HPLC-DAD analysis,and their n-octanol / water partition coefficients were measured by shaking flask combined with HPLC-DAD. RESULTS The equilibrium solubilities of polydatin and resveratrol were found in their maximal values at p H 6. 8,while that of emodin was at p H 6. 5. the scopes of n-octanol / water partition coefficients of polydatin,resveratrol and emodin in different p H buffer solutions were within the ranges of 0. 84- 1. 01,2. 34- 2. 89,and 0. 97- 1. 27,respectively. And their lg Pappin n-octanol / water solvent were shown to be 0. 98,2. 69,and 0. 99,respectively. CONCLUSION Unlike resveratrol,polydatin and emodin have good absorption under physiological conditions in the gastrointestines.
