丹参酮ⅡA磺酸钠对大鼠心肌缺血再灌注脂质氧化的影响 被引量:3

摘要 目的:探究丹参酮ⅡA磺酸钠提前干预对大鼠缺血再灌注损伤心肌系统脂质超氧化起到的保护功效。方法:30只雄性SPF级SD大鼠,平均分为伪手术组、缺血再灌注组(对照组)、丹参酮ⅡA磺酸钠提前干预组(丹参酮ⅡA组),各组数量均为10只。丹参酮ⅡA组于结扎前2h自大鼠尾静脉注射丹参酮ⅡA磺酸钠液1.5mg/kg,伪手术组和对照组结扎前2h静脉注射等量生理盐水。除了伪手术组之外,另外两组结扎左冠状动脉前降支,让心肌处于缺血状态,维持20min,接着再灌注维持1h。再灌注1h后处死大鼠,速取大鼠心肌组织,用ELISA法测定心肌超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)活性和丙二醛(MDA)含量;观察心肌组织病理学变化。结果:相较于伪手术组,对照组心肌组织LDH、CK-MB及SOD活性下降(P<0.05),MDA含量升高(P<0.05);相较于对照组,丹参酮ⅡA组心肌组织LDH、CK-MB及SOD活性升高(P<0.05),MDA含量降低(P<0.05)。结论:丹参酮ⅡA磺酸钠提前干预能减轻大鼠缺血再灌注心肌组织脂质超氧化,对保护心肌系统有较好的功效。 Objective:To investigate early intervention of the sodium tanshinoneⅡA on protection effect of myocardial ischemia-reperfusion injury lipid superoxide. Methods:30 SPF SD male rats were divided into sham group, ischemia-reperfusion group (control group), sodium tanshinone ⅡA advance intervention group (Tan ⅡA group);each group was 10 cases. Tan ⅡA group was given tail vein injection of sodium tanshinoneⅡA by 1.5mg/kg. Except the fake-surgery group, in the other two groups, the left anterior descending coronary artery was ligated, so in myocardial ischemia for 20 minutes, followed by reperfusion for 1 hour. Measuring myocardial superoxide dismutase (SOD) by ELISA, lactate dehydrogenase (LDH), creatine kinase (CK-MB) activity and malondialdehyde (MDA) content;myocardial histopathological changes. Results:Compared to the fake control group, in the control group, myocardial tissue LDH, CK-MB and SOD activity decreased (P〈0.05), MDA content increased (P〈0.05);Compared to the control group, in the Tan ⅡA group myocardial tissue LDH, CK-MB and SOD activity increased (P〈0.05);MDA decreased (P〈0.05). Conclusion:Early intervention of TanⅡA sodium can alleviate ischemia-reperfusion myocardial tissue lipid superoxide, with better protecting effects.
