摘要
[目的 ]应用引进的Va间接培养方法、传统的丙酮酸钠、L-J培养基直接培养方法对牛结核分枝杆菌生长时间进行对比试验。[方法 ]将牛型结核分枝杆菌接种于含0.1%琼脂Va培养基中预培养24 h,转接到含0.3%琼脂Va培养基中培养6 d后分别转接到丙酮酸钠培养基和L-J培养基上(间接法);同时牛型结核分枝杆菌接菌于丙酮酸钠培养基和L-J培养基培养,在37℃培养箱内孵育(直接方法),每天观察,并记录出现菌落时间。[结果]接种Va培养基的菌株分别转接丙酮酸钠培养基和L-J培养基后第3 d可见菌落,第12 d培养基斜面可见典型的牛型结核分枝杆菌菌落;而直接接种于丙酮酸钠培养基与L-J培养基后分别于第26 d和第31 d可见典型的牛型结核分枝杆菌菌落。[结论 ]引进的Va牛型结核分枝杆菌间接培养方法可以缩短菌落培养时间,为牛型结核杆菌病提供早期诊断依据。
[Objective] To apply the introduction of Va medium composition indirect method and the traditional culture medium of sodium pyruvate,L-J medium cultivation method by direct methods,to compare the bovine mycobacterium tuberculosis growth time. [Method]The standard strains,which were processed with pre-culture in 0.1% agar Va media during 24 h and incubated in 0.3% agar Va media for 6 days at 37℃,were transferred into sodium pyruvate medium and L-J medium respectively. Meanwhile the standard strains were directly inoculated into sodium pyruvate medium and L-J medium. And then via the incubation at 37,it was observed that the examination was done daily until growth of colonies. [Result]Growth of mycobacteria cultures of Mycobacterium bovis in sodium pyruvate medium and L-J medium was observed on the 3th day by the indirect method,and growth of the typical colonies were observed on the 12 th day in the two mediums. However,growth of the colonies of mycobacteria in the sodium pyruvate medium and the L-J medium did on the 26 th day and the 31 th day through direct methods respectively. [Conclusion]Introduction of the indirect method can shorten the growth time of the colonies,and provide early diagnosis basis for Mycobacterium bovis.
