抗山羊痘病毒P32蛋白杂交瘤细胞的构建及其单克隆抗体的制备 被引量:3

摘要 本研究旨在以纯化的山羊痘病毒P32蛋白为免疫原,构建杂交瘤细胞株,制备P32蛋白单克隆抗体。用纯化的P32蛋白免疫BALB/C小鼠后,按常规方法进行细胞融合,构建抗山羊痘病毒P32蛋白的杂交瘤细胞,制备P32蛋白单克隆抗体,获得4株能识别重组蛋白且特异性强无交叉反应的杂交瘤细胞株,腹水抗体效价均在10^5以上。山羊痘病毒P32蛋白杂交瘤细胞株的构建及单克隆抗体的制备为进一步开展检测方法研究提供了材料。 The study was aimed to construct hybridoma cell line by using purified goat poxvirus P32 protein as immunogen for preparation of P32 protein monoclonal antibody. After BALB/C mice were immunized with purified P32 protein of goat poxvirus,cell fusion was carried out according to the routine method,resulting in 4 hybridoma cell lines,which could stably secrete P32 protein monoclonal antibody with good specificity. The ascites antibody titers were higher than 10-5. Construction of goat poxvirus P32 protein and preparation of monoclonal antibodies provided material for further research on the virus.
