摘要
对苏云金杆菌Bt02进行发酵条件的优化研究,通过碳源和氮源单因素试验及正交试验,选出最佳碳、氮源及配比,对p H值、发酵温度和转速进行了研究,结果表明,碳源选用麦芽糊精,氮源选择脱脂豆粉,初始p H值8.0,温度30℃、转速为200 r/min为最佳发酵条件,菌数提高到8×109cfu/m L。相对于原配方的4.3×109cfu/m L,提高了1.86倍。
The fermentation conditions of Bacillus thuringiensis Bt02 were optimized and preliminary tested.Through single factor test of carbon and nitrogen source and orthogonal test, the best ratio of carbon, nitrogen source were selected, pH value, fermentation temperature and rotational speed were also studied. The results showed that the best carbon source was malt dextrin, and the best nitrogen source was defatted soybean meal. The initial pH 8.0, temperature 30℃, the speed of 200 r/min was the best fermentation conditions and the microbial content was up to 8× 10^9 cfu/mL.Relative to the original recipe whose microbial content was 4.3× 10^9 cfu/mL, the count of new recipe increased 1.86 times.
