机采血小板低温冰冻保存与常规液态保存的主要参数比较 被引量:3

摘要 目的对照常规液态保存条件下,实际观察血小板计数(PLT)血小板平均体积(MPV)、血小板分布宽度(PDW)、p H值、低渗休克反应(HSR)和CD62p参数,作为血小板冰冻保存的主要质量监控数据。方法采用Trima5.0血细胞分离机采集血小板32个治疗单位(血小板数≥2.5×1011/L,容量250~300 ml),22℃振荡保存96 h后,采用无菌导管接口机留样10 ml为新鲜组,保存袋中加入终浓度5%的二甲亚砜(DMSO)低温保护剂,采用无菌导管接口机留样10 ml。冰冻样管置-80℃冰冻保存10个月,使用时42℃水浴溶解。结果相比较96 h常规保存条件下,-80℃冰冻保存10个月后解冻血小板计数的差异无统计学意义(P〉0.05),MPV、PDW的差异有统计学意义(P〈0.05),p H值的差异无统计学意义(P〉0.05),HSR、CD62p的差异有统计学意义(P〈0.05)。结论相对于机采新鲜血小板,冰冻血小板的活力有所下降,但活化程度有所提高。 Objective To compare platelet count,MPV,PDW,PH value,hypotonic shock response( HSR) and CD62 P in fresh and cryopreserved apheresis platelets,that are referred to as the main quality parameters of platelets in cryopreservation and apheresis. Methods Thirty-two bags of platelets( ≥2. 5 × 1011/ L,250 ~ 300 ml / bag) were collected by Trima5. 0 blood cell separator,and an aliquot of 10 ml platelets from each bag was taken as fresh group after 96 hours preserved at 22℃ and frozen group after 10 months preserved with 5% dimethyl sulfoxide at- 80℃,which were thawed at 42℃ in water bath before using. Results No significant difference was seen in platelet counts and PH value( P〉0. 05),but significant differences were presented between MPV,PDW,HSR and CD62p( P〈0. 05) compared to fresh group. Conclusions Compared to fresh apheresis platelets,the activity of cryopreserved platelets was decreased but activation seems to be elevated.
