基于SRAP的15种鸢尾的遗传多样性和亲缘关系分析 被引量:1

摘要 为探讨北方地区15种鸢尾的遗传多样性和亲缘关系,采用SRAP分子标记的方法对鸢尾遗传多样性的亲缘关系进行评价。结果表明:利用优化的SRAP-PCR反应体系和筛选出的6对引物组合对15种鸢尾扩增,共得到207条清晰稳定的条带,片段大小是50~1500bp,多态位点百分比为100%。多态信息含量为0.3976,遗传多样性参数分别为Na=2.000,Ne=1.6947±0.2416,H=0.3967±0.0941,I=0.5816±0.1065,说明15种鸢尾在种水平上有较高的遗传多样性,并有较大的遗传分化。鸢尾的亲缘关系通过UPGMA聚类分析和主成分分析两种方法进行,在遗传相似性系数为0.589时,15种鸢尾被分成六大类群。该研究为鸢尾属植物种质资源的分类、鉴定、保护及新品种选育等方面提供参考。 The genetic diversity and relationship among 15 Iris species in Northern China were analyzed by SRAP(sequence related amplified polymorphism, SRAP). 207 clear and stable bands were generated by the improved SRAP-PCR system and 6 primer groups screened, ranging in size from 50 to 1500 bp, with 100% the percentage of polymorphic bands. The polymorphism information content(PIC) value was 0.3976, and the value of genetic diversity parameters were Na=2.0000±0.0000, Ne=1.6947±0.2416, H=0.3967±0.0941,I =0.5816 ±0.1065, respectively, which indicated that the genus of Iris was a great genetic diversity at inter-specific level. The genetic relationship was analyzed by UPGMA clustering analysis and the principle coordinate analysis. Fifteen collections were separated into6 groups at a genetic similarity coefficient of 0.589. The objective of this research was to provide scientific references for identification,classification, conservation and breeding.
