摘要
为探讨东方百合新品种Double surprise的组培快繁技术,以其花瓣为初级外植体,以初级外植体诱导形成的无菌苗鳞片和叶片作为次级外植体,分别探讨了外植体诱导、不定芽增殖、组培苗生根以及炼苗移栽的较优条件。结果表明:花瓣诱导不定芽的较优培养基为MS+6-BA2.0mg·L-1+NAA0.3mg·L-1,诱导率可达86.67%,诱导系数为2.73;MS+TDZ2.0mg·L-1+NAA0.5mg·L-1为诱导无菌苗鳞片和叶片直接产生不定芽的较优培养基,不定芽诱导率分别为88.89%和91.11%,诱导系数分别为6.67和5.67;MS+PIC1.0mg·L-1+TDZ0.5mg·L-1为无菌苗鳞片间接诱导愈伤组织的较优培养基,愈伤组织诱导率和分化系数分别为90.00%和3.27;MS+PIC 1.0 mg·L-1+TDZ 0.1 mg·L-1为无菌苗叶片间接诱导愈伤组织的较优培养基,愈伤组织诱导率和分化系数分别为93.33%和3.70;较优的不定芽增殖培养基为MS+6-BA 2.0 mg·L-1+NAA 0.3 mg·L-1;不定芽生根的较优培养基为1/2MS+IBA 1.0 mg·L-1;草炭∶蛭石∶珍珠岩(体积比)=1∶1∶1的混合基质为Double surprise较优移栽基质,成活率可达88%。
In order to study the technology of tissue culture and rapid propagation on oriental lily, the petals segments of double surprise were used as original explants, the scales and leaves of in vitro stock bulblets obtained from the first explants as the secondary explants, we investigated the optimum conditions of shoots induction and differentiation, roots inducement and transplanting. The results showed that the best shoots induction medium for petals was MS+ 6-BA 2.0 mg·L-1+ NAA 0.3 mg·L-1, in which the average induction rate was 86.67% and the induction factor was 2.73. For scales and leaves of in vitro stock bulblets,the optimum shoots inducement medium was MS+TDZ 2.0 mg·L-1+ NAA 0.5 mg·L-1, in which the average induction rate was 88.89%and 91.11%, respectively. While the best induction medium for callus induction was MS + PIC 1.0 mg·L-1+ TDZ 0.5 mg·L-1and MS +PIC 1.0 mg·L-1+ TDZ 0.1 mg·L-1, in which the induction rate of callus was 90.00% and 93.33%, respectively. The most suitable medium for shoots propagation was MS + BA 2.0 mg·L-1+ NAA 0.3 mg·L-1, and 1/2MS + IBA 1.0 mg·L-1was best for rooting. The regenerated plantlets survived with highest rate of 88.00% in substrate of peat∶vermiculite∶ perlite(volume ratio) = 1∶1∶1.
