摘要
目的探究"气阴两虚型心衰病证结合模型"的构建与评价。方法首先探究过量异丙肾上腺素(ISO)造成心衰小鼠模型的方案,以超声心动、脏器指数、病理切片等明确ISO剂量;在此基础上,联合剥夺睡眠的方式,构建气阴两虚型心衰病证结合小鼠模型,并通过观察宏观体征、图像处理量化分析、旷场实验等评价模型。结果递减剂量组(第1天20 mg/kg,第2天10 mg/kg,第3~14天5 mg/kg)相比恒定剂量组(ISO 5 mg/kg连续30 d)心衰更显著;剥夺睡眠组相比单纯注射ISO组气阴两虚程度更明显。结论联合皮下注射异丙肾上腺素和小站台水环境法剥夺睡眠可构建气阴两虚型心衰病证结合小鼠模型。
Objective: To explore the construction and evaluation of the Qi and Yin deficiency pattern of heart failure mouse model. Methods: The programme of excess isoproterenol mouse model with heart failure was ex- plored,valued by ultrasonic cardiography,organ index and pathology. On the basis of the heart failure mouse model,sleep deprivation way was combined to build the Qi and Yin deficiency pattern of heart failure rat model and the model was evaluated by observing macroscopic signs, open-field test, and image processing quantification analysis. Results: The dose decreasing group (the 1st day 20 mg/kg,the 2nd day 10 mg/kg,from 3rd to 14th day 5 mg/kg) had a severer heart failure compared with the dose constant group, (ISO 5 mg/kg for 30 days) and the group of sleep deprivation had a higher degree on Qi and Yin deficiency. Conclusion: Subcutaneous injection of isoproterenol combined with small platform water environment to perform sleep deprivation can build Qi and Yin Deficiency type of mouse model with heart failure disease and syndrome.
