活血通络解毒方对脑缺血Wistar大鼠炎症细胞因子TNF-α、IL-1β、IL-6的影响 被引量:4

摘要 目的研究活血通络解毒方对脑缺血Wistar大鼠神经缺损功能评分及肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的影响。方法 60只雄性Wistar大鼠随机分成假手术组,模型组,活血通络解毒方低剂量组、中剂量组、高剂量组5组。大鼠大脑中动脉栓塞模型造模成功24 h、7 d、14 d后,各组给予相应处理,分别分析比较检查各组神经功能评分情况及TNF-α、IL-1β、IL-6水平。结果假手术组无大鼠死亡,模型组死亡2只,分别死于术后第2日、第8日,活血通络解毒方高剂量组死亡2只,分别死于术后第6日和第9日。活血通络解毒方中、低剂量组未见动物死亡。造模后各组大鼠神经功能缺损评分与假手术组比较均升高(P<0.01)。各治疗组在治疗后其评分与模型组比较均降低(均P<0.05),且中剂量组神经功能缺失评分第7 d明显低于模型组(P<0.01)。治疗中剂量组神经功能缺失评分从术后第7日、14日均低于高剂量组和低剂量组(P<0.05),高剂量组和低剂量组评分差别不大(P>0.05)。造模后24 h各组大鼠血清TNF-α、IL-1β、IL-6水平与假手术组比较均明显升高(均P<0.05),而模型组、高、中、低剂量组间比较则差别不大(均P>0.05)。术后2周假手术组、模型组TNF-α、IL-1β、IL-6水平与本组术后24 h比较差别不大(均P>0.05);术后2周高、中、低剂量组TNF-α、IL-1β、IL-6水平与本组术后24 h比较均下降(均P<0.05),且中剂量组下降最明显(P<0.05)。术后2周高、中、低剂量组TNF-α、IL-1β、IL-6水平与模型组同期比较均下降(均P<0.05)。结论活血通络解毒方可改善缺血性Wistar大鼠神经功能缺损评分及其炎症因子TNF-α、IL-1β、IL-6水平,这可能是干预脑梗死的机制之一。 Objective: To Study the efficacy of Huoxue tongluo jiedu Decoction on inflammatory cytokine TNF-α,IL-1β,IL-6 of rats with cerebral ischemia Wistar. Methods: Sixty Wistar Mice were randomly derided into sham-operated group, the model group, group with low level of Huoxue tongluo jiedu Decoction, group with mod- erate level, and group with high level. The middle cerebral artery occlusion reperfusion model in rats was built ac- cording to Longa. 24h after finishing modeling and 7 and 14 days after the treatment, the level of TNF-c~, IL-113 and IL-6 of each group were detected,and the neurobehavioral change was observed. Results : At the end of the experiment,there were no rats dead in sham operation group and group with low level,2 in the model group on 2nd and 8th day respectively after operation, 2 in group with high level on 6th and 9th day respectively after op- eration. After finishing modeling,compared with sham operation group, Neurological deficit scores of other groups significantly increased,with statistical significance (P 〈 0.01). Compared with the model group,the situation of neural function was improved, and the level of TNF-α,IL-1β and IL-6 of each group increased (P〈 0.05). After 14 days' treatment,the level of TNF-α,IL-1β and IL-6 of each group decreased,among which,the group with moderate dose decreased the most (P〈 0.05). Conclusion: Huoxue tongluo jiedu Decoction can improve Neuro- logical deficit scores and decrease the level of TNF-α,IL-1β,IL-6,which may be one of the mechanisms of cerebral infarction.
