miR-487a在肾透明细胞癌中的表达及临床意义

摘要 目的探讨miRNA-487a在肾透明细胞癌(ccRCC)中的表达及临床意义。方法收集2011年1月至2012年12月在南京军区南京总医院泌尿外科手术切除的45例配对ccRCC组织及癌旁正常肾组织,常规提取45例新鲜ccRCC与癌旁组织中总RNA,应用荧光定量反转录-聚合酶链反应法检测标本中miRNA-487a的表达状况;分析miRNA-487a表达差异与临床病理特征间的关系。结果与癌旁组织(1.00)比较,miRNA-487a在ccRCC表达水平为(3.30±1.03),明显上调(P<0.05)。miRNA-487a的表达在不同肿瘤浸润程度及Fuhrman分级间差异有统计学意义(P<0.05),而与年龄、局部淋巴结转移和远处转移均无关(P>0.05)。结论 miRNA-487a在肾透明细胞中表达上调,可能为促进ccRCC的发生、发展的因素之一。 Objective To detect the expression and clinical significance of miRNA-487 a in clear cell renal cell carcinoma( ccRCC). Methods ccRCC and corresponding adjacent tissues of 45 patients admitted to the Department of Urology,Nanjing General Hospital of Nanjing Military Command from Jan. 2011 to Dec.2012 were collected and the total RNA was extracted routinely. The miRNA-487 a expression was detected by using fluorescent quantitative reverse transcription-polymerase chain reaction. The correlations of miRNA-487 a expression with the clinicopathologic characteristics was analyzed. Results The miRNA-487 a expression level of ccRCC tissue was( 3. 30 ± 1. 03),which was significantly higher than the adjacent tissue( P〈0. 05). The expression of miRNA-487 a had statistically significant difference between different invasion degrees and Fuhrman gradings( P〈0. 05),but showed no significant correlations with the patients' age,histological type of the tumor,lymph node metastasis and distal metastasis( P〈0. 05). Conclusion The up-regulated expression level of miRNA-487 a in ccRCC is correlated with invasion degree and Fuhrman grading,and it may participate in the pathogenesis and development of ccRCC.
