Hydrolysate Alalyze and Enzymatic Saccharification Conditions Optimization of Reed by Steam Explosion Pretreatment

摘要 芦苇(Phragmites australis)是一种有潜力的能源作物,为优化芦苇酶解糖化工艺,应用Plackett-Burman试验设计筛选影响芦苇酶糖化的重要参数,通过Box-Behnken设计确定重要参数的最佳水平,应用高效液相色谱仪(HPLC)对糖化过程中的单糖种类及含量进行分析.结果表明,影响芦苇酶糖化的重要参数是H2SO4浓度、Tween-80和MnSO4浓度,最佳工艺参数为H2SO4浓度0.88%、表面活性剂Tween-80添加量0.61%、MnSO4添加量0.26%,在此条件下,8~10 h可以完成糖化,总还原糖浓度达到45.68 mg/mL,同时验证了数学模型的有效性, 液相分析表明糖化液中主要的糖种类为葡萄糖和木糖, 其含量分别为21.36、16.62 mg/mL,阿拉伯糖、纤维二糖和半乳糖含量较少. Reed (Phragmites australis)was a kind of alternative raw material for bio-ethanol production. The important parame-ters of influencing enzymatic sacharification for reed were screened by Plackett-Burman design. Furthermore, based on Box-Behnken design, the enzymatic saccharification conditions of reed by steam explosion pretreatment were optimized by response surface methodology, and a mathematial model of a second order quadratic equation was developed for reducing sugar conen-tration. The hydrolysate was alalysed by HPLC. The resutlts suggested that the three important parameters included dilute sul-phuric acid, Tween-80 and manganese sulfate concentration. The optimized technologial parameters were as follows: dilute sulphuric acid concentration 0.88%,、Tween-80 concentration 0.61%, manganese sulfate contration 0.26%. The reducing sugar concentration was up to 45.68 mg/mL. The results verified the validity of the mathematical model. The concentrations of glu-cose and xylose were 21.36 mg/mL and 16.62 mg/mL, respectly. Contents of arabinose, cellobiose and galactose were very low.
