应用多功能荧光酶标仪分析盐酸阿霉素含药量 被引量:4

摘要 目的建立应用多功能荧光酶标仪对盐酸阿霉素(多柔比星)含药量直接进行荧光分析的方法。方法在体系为0.050 mol/L氨丁三醇(p H 6.30)条件下设定激发波长和发射波长分别为495、595 nm,应用多功能酶标仪测定盐酸阿霉素荧光值。结果盐酸阿霉素为0.78~25.00μM时荧光强度呈良好线性(R2=0.979),加样回收率达97.97%以上。该法用于测定SD大鼠血样中盐酸阿霉素时回收率为98.30%~98.80%。结论该法具有较好的准确度与灵敏度,适用于盐酸阿霉素含药量测定及在血样中的分析测定。 Objective To establish a direct fluorometric analytical method by using the multifunctional fluorescent en-zyme-labeled instrument for detecting the doxorubicin hydrochloride content. Methods Under the condition of the system as0.050 mol/L tromethamine,the excitation wavelength and emission wavelength were set as 495 nm and 595 nm,the multifunctional fluorescent enzyme-labeled instrument was used to detect the fluorometric value of doxorubicin hydrochloride. Results The fluorescence intensity of doxorubicin hydrochloride had a good linearity when doxorubicin was 0.78-25.00 μM(r2=0.979),the recovery rate was above 97.97%. The recovery rate was 98.30%-98.80% when detecting the recovery rate of doxorubicin in SD rat blood sample. Conclusion The established method has better accuracy and sensitivity,and is suitable for the determination of doxorubicin hydrochloride content and the analytic determination of blood sample.
