Effect of epidermal growth factor receptor on migration and invasion ability of endometrial carcinoma cells

摘要 目的选取表皮生长因子受体(EGFR)低表达的子宫内膜癌Ishikawa细胞和经转染过表达EGFR的Ishikawa-p LWERNL细胞,研究EGFR对子宫内膜癌细胞迁移能力和侵袭能力的影响。方法体外迁移试验比较转染前后2种子宫内膜癌细胞的迁移能力。体外侵袭试验检测转染前后2种子宫内膜癌细胞的侵袭能力。蛋白印迹法比较转染前后2种子宫内膜癌细胞基质金属蛋白MMP9、MMP2表达水平的差异。结果和转染前的Ishikawa细胞相比较,经转染稳定过表达EGFR的Ishikawa-p LWERNL细胞迁移能力更强,差异有统计学意义(t=6.148,P<0.01)。转染后,稳定过表达EGFR的Ishikawa-p LWERNL细胞侵袭能力增强,差异有统计学意义(t=10.028,P<0.01)。和EGFR低表达的Ishikawa细胞相比较,EGFR高表达的Ishikawa-p LWERNL细胞基质金属蛋白酶MMP9、MMP2的表达水平较高,差异有统计学意义(t值分别为6.934,5.748,P值均<0.05)。结论 EGFR过表达可增强子宫内膜癌细胞的迁移能力和侵袭能力。 Objective To select endometrial carcinoma Ishikawa cells with low expression of epidermal growth factor receptor( EGFR) and Ishikawa- p LWERNL cells with overexpression of EGFR after transfection,research the effect of EGFR on migration and invasion ability of endometrial carcinoma cells. Methods In vitro migration test was used to compare migration ability of endometrial carcinoma cells before and after transfection. In vitro invasion test was used to detect the invasion ability of endometrial carcinoma cells before and after transfection. Western blot was used to compare the expression levels of matrix metalloproteinase 9( MMP- 9) and MMP- 2 in the two kinds of endometrial carcinoma cells before and after transfection. Results Compared with Ishikawa cells before transfection,the migration ability of Ishikawa- p LWERNL cells was stronger( t = 6. 148,P〈0. 01). After transfection,invasion ability of Ishikawa- p LWERNL strengthened significantly( t = 10. 028,P〈0. 01). Compared with Ishikawa cells,the expression levels of MMP- 9 and MMP- 2 in Ishikawa-p LWERNL cells increased significantly( t = 6. 934,P〈0. 05; t = 5. 748,P〈0. 05). Conclusion Overexpression of EGFR can enhance migration and invasion ability of endometrial carcinoma cells.
