摘要
目的采用斑马鱼焦虑行为学模式,研究菊葛天麻颗粒(JGTM)单用、与丁螺环酮(Bus)合并应用抗焦虑作用。方法 AB系野生斑马鱼12月龄,实验分为两个系列:单独用药与两药合并应用。单独用药:斑马鱼30条,随机分为5组(n=6),模型对照组(鱼在100 m L水体中暴露3 min),Bus组(鱼在同体积6.25mg/L Bus的水体中暴露3 min),JGTM低、中、高剂量组(鱼分别在同体积0.5,1.0,2.0 g/L JGTM的水体中暴露3 min);合并用药:斑马鱼30条,随机分为5组(n=6),模型对照组和Bus组(同单独用药系列),JGTM与Bus合并用药组(鱼在同体积6.25mg/L Bus及0.5,1.0,2.0 g/L JGTM的水体中暴露3 min);以上试验组别均给药一次。用斑马鱼行为视频跟踪分析系统自动记录斑马鱼行为参数,比较大运动次数、时间、距离等变化。结果 JGTM 1.0 g/L为抗焦虑作用的有效剂量,该剂量与Bus 6.25 mg/L合并应用,斑马鱼大运动次数、时间、距离等行为学参数均有显著性变化(P<0.01);与Bus 6.25 mg/L组比较,大运动的时间、距离显著增加(P<0.05)。结论 JGTM具有一定的抗焦虑作用,与Bus合并应用具有抗焦虑协同作用。
Objective To study the pharmacodynamic effects of Juge Tianma Granules (JGTM) used alone or combined with buspirone (Bus) on anxiety-like behavior of zebrafish. Methods AB zebrafish of 12 months old were used in the experiment composed of two series, in which JGTM were used alone or combined with Bus. When used alone, 30 zebrafish were divided into 5 groups (n=6): control group (zebrafish were exposed to 100 mL water for 3 rain), Bus group (zebrafish were exposed to 100 mL of 6.25 mg/L Bus solution for 3 rain), JGTM 0.5, 1.0, 2.0 g/L groups (zebrafish were exposed to 100 mL of 0.5, 1.0 or 2.0 g/L JGTM solution for 3 min). When combined with Bus, 30 zebrafish were divided into 5 groups (n=6): control group, Bus group, JGTM 0.5, 1.0, 2 .0 g/L and Bus groups (zebrafish were exposed to 100 mL of 0.5, 1.0 or 2.0 g/L JGTM and 6.25 mg/L Bus solution for 3 rain). Every above test group was treated once only. With the video record and track system, the behavior was recorded and quantified automatically. Results 1.0 g/L is the effective anxiolytic dose of JGTM; 1.0 g/L JGTM combined with 6.25 mg/L Bus can increase larct, lardur and lardist of zebrafish significantly (P〈0.01). Compared with Bus treated group, the lardur and lardist of zebrafish increase significantly (P〈0.05). Conelusion JGTM Granules have anxiolytic effects, as well as synergistic effect when combined with Bus.
