活血药对肺纤维化模型大鼠肺组织超氧化物歧化酶和谷胱甘肽表达的影响 被引量:6

摘要 目的:探讨活血化瘀类中药对博来霉素诱导的大鼠肺纤维化模型肺组织病理变化及血清中超氧化物歧化酶(SOD)、谷胱甘肽(GSH)表达水平的影响。方法:SPF级健康雄性SD大鼠32只,随机数字表法分为四组:对照组、模型组、中药组、泼尼松组,采用气管内滴入博来霉素的方法制作肺纤维化模型。造模后第2天起,对照组及模型组予以生理盐水灌胃,中药组以丹参、川芎水煎液灌胃,泼尼松组以泼尼松混悬液灌胃。28 d后处死各组大鼠,光镜下观察各组大鼠肺组织病理变化,化学荧光法测定大鼠血清中SOD、GSH表达水平。结果:模型组大鼠血清SOD含量较其他组明显降低(P<0.01),中药组较对照组略低(P>0.05),中药组较泼尼松组无明显差异;模型组大鼠血清GSH水平较其他三组明显降低(P<0.01);中药组、泼尼松组较对照组GSH水平偏低(P<0.01)。结论:活血化瘀中药丹参和川芎可以提高肺纤维化模型大鼠血清SOD、GSH,减轻博莱霉素诱导的肺泡炎症和肺纤维化程度。 Objective:To discuss the effect of herbs promoting blood circulation to remove blood stasis of Chinese traditional medicine for the expression of superoxide dismutase(SOD) and glutathione(GSH) in rats’ lung tissue with Pulmonary fibrosis which induced by bleomycin. Methods:32 SPF SD rats were divided into control group,model group,Chinese herbs group and prednisone group randomly,8 rats for each group. Bleomycin was instillated into intratracheal to make rat model of pulmonary fibrosis. The control group was given physiological saline by gavage,the Chinese herbs group were given Danshen,Chuanxiong decoctio,and the rednisone group were given Prednisone after 2 days later. All the rats were sacrificed after 28 days ,and observed the pathological changes of lung tissue under the microscope,determinate SOD,GSH level in serum by chemiluminescence,the expression. Results:The content of SOD of the model group is less than those of other three groups(P〈0.01). The Content of SOD in Chinese herbs group has no statistical difference com-pare with the control group and the prednisone group (P〉0.05);The content of GSH of the model group re duce than those of other three groups(P〈0.01);the Chinese herbs group and the prednisone group reduce than the control group(P〈0.01). Conclusions:Promoting blood circulation and removing blood stasis herbs of traditional Chinese medicine can improve pulmonary fibrosis model of rat serum SOD and GSH ,induce alve-olitis and pulmonary fibrosis degree.
