摘要 目的:检测不同基因型间T细胞免疫球蛋白黏蛋白域蛋白-1(Tim-1)在系统性红斑狼疮患者和健康对照组血清中的表达浓度及其基因第4外显子插入/缺失多态性,分析其与系统性红斑狼疮的临床意义。方法:采用ELISA方法测定Tim-1蛋白在血清中的浓度;PCR反应检测Tim-1的第4外显子插入与缺失多态性,计算不同基因型的表达频率。结果:系统性红斑狼疮患者和健康对照组血清中Tim-1蛋白的浓度分别是:(268.063±256.752)和(60.327±89.323)pg/ml,两组之间的差异有统计学意义;SLE患者Tim-1的第4外显子缺失/插入杂合子与缺失/缺失纯合子的Tim-1蛋白的浓度分别为(187.630±244.688)pg/ml和(311.356±272.119),健康对照组Tim-1的第4外显子缺失/缺失纯合子与缺失/插入杂合子的Tim-1蛋白的浓度分别为(72.285±107.827)和(43.358±41.669)pg/ml,两组之间无显著性差异。对照组的Tim-1的第4外显子缺失/缺失纯合子,缺失/插入杂合子,插入/插入纯合子基因型频率分别是0.632,0.318,0.071;系统性红斑狼疮患者相应的基因型频率是0.656,0.324,0.030。两组之间无显著性差异。结论:Tim-1蛋白在系统性红斑狼疮患者血清中的表达浓度升高,与系统性红斑狼疮的发生呈正相关性,但第4外显子插入与缺失的基因多态性与系统性红斑狼疮无相关性,该突变与Tim-1蛋白在血清中的表达无相关性。 Objective:To detect the expression concentration of T cell immunoglobulin domain and mucin domain protein-1(Tim-1)of different genotypes concentration in the serum of patients with systemic lupus erythematosus and healthy control group and insertion and deletion polymorphisms of exon 4and analyze its clinical significance to systemic lupus erythematosus.Methods:Detect the concentration of Tim-1protein in serum according to ELISA;determine insertion and deletion polymorphisms of Tim-1exon 4according to PCR,and work out the expression frequency of various genotypes.Results:The concentration of Tim-1in the serum of patients with SLE and healthy control group were(263.083±276.953)and(58.527±92.424)pg/ml,the difference between the two groups was statistically significant;the concentration of Tim-1of patients with deletion in exon 4and insertion of heterozygote and deletion in exon 4and insertion of homozygote was(187.630±244.688)pg/ml and(311.356±272.119)respectively;while in the healthy control group,the concentration of Tim-1with deletion in exon 4and deletion of homozygote and deletion in exon 4and insertion of heterozygote was(72.285±107.827)and(43.358±41.669)pg/ml respectively,there was no significant difference between the 2groups.The Genotype frequency of Tim-4with deletion in exon 4and deletion in homozygote,deletion in exon 4and insertion of heterozygote and insertion and insertion of homozygote was 0.632,0.318,0.071 and the corresponding frequency of patients with SLE was 0.656,0.324,0.030,there was no significant difference between the 2groups.Conclusion:The increase in the expression concentration of Tim-1in the serum of patients with SLE is positively relate to the incidence of systemic lupus erythematosus,but there is no correlation between insertion and deletion polymorphisms of exon 4insertion and systemic lupus erythematosus,the mutation is not relative to the expression of Tim-1in serum.
