摘要
目的研究氟离子导入对猪骨源性羟基磷灰石(porcine hydroxyapatite,PHA)蛋白吸附行为的影响。方法制备PHA将PHA骨块分别浸泡于氟离子浓度为0.25 mol/L、0.50 mol/L、0.75 mol/L、1.00 mol/L的氟化钠溶液中24 h;取出骨块煅烧、冲洗、干燥后得到4种渗氟浓度的氟化猪骨源性羟基磷灰石(fluorinated porcine hydroxyapatite,FPHA)骨块(分别用FPHA0.25、FPHA0.50、FPHA0.75、FPHA1.00代表)。检测PHA和4种渗氟浓度的FPHA对牛血清白蛋白(bovine serum albumin,BSA)单种蛋白的吸附能力;采用凝胶电泳法检测PHA和4种渗氟浓度的FPHA对BSA和溶菌酶(lysozyme,LSZ)的双蛋白竞争吸附能力。结果 FPHA较PHA有更强的蛋白吸附能力且随氟离子导入浓度增加,FPHA对BSA的吸附能力逐渐增强;在双蛋白竞争吸附情况下,FPHA和PHA对BSA的吸附与对BSA单种蛋白时的吸附规律一致,而对LSZ的吸附则随氟离子导入浓度的增加而减少。结论氟离子导入增强了PHA吸附BSA的能力;但在双蛋白竞争条件下,吸附LSZ的能力则下降。
Objective To explore the effects of fluoride iontophoresis on protein adsorption of porcine derived Hydroxyapatite(PHA).Methods PHA bones were prepared and immersed in sodium fluoride solution with indicated concentrations(F:0.25,0.50,0.75,1.00 mol/L) for 24 h;Then the bones were removed,treated by calcinated,washed,and dried subsequently;4 subgroups of fluorinated porcine hydroxyapatite(FPHA) bones were finally harvested,representing as FPHA0.25,FPHA0.50,FPHA0.75,and FPHA1.00,respectively.Bovine serum albumin(BSA) was used to determine the single protein adsorption of PHA and the 4 groups of FPHA,while BSA and lysozyme(LSZ) were used to test the dual- protein competitive adsorption by gel electrophoresis(SDS-PAGE).Results It showed stronger protein adsorption of FPHA compared with PHA,and BSA adsorption of FPHA was enhanced with the increase of fluorine concentration.Adsorption of BSA of FPHA and PHA was similar with the single protein adsorption in dual protein competitive adsorption,however,LSZ adsorption was reduced with the increase of fluorine concentration.Conclusion Fluoride iontophoresis may enhance BSA adsorption of PHA while reduce LSZ adsorption in dual-protein competition.
