摘要
磷脂酶D可以催化合成磷脂酰丝氨酸,为了得到高活力的磷脂酶D,采用蛋黄平板法从油脂厂区的土壤中筛选出产磷脂酶D的菌株X1。通过摇瓶培养,用TLC薄层层析法测定酶活力,达到2 023U/L。对发酵培养基组分及发酵条件进行优化,结果表明,葡萄糖为碳源10g/L,蛋白胨为氮源15g/L,发酵温度为30℃,发酵液初始pH为7,摇床转速为180r/min的发酵条件,酶活力达到最大,为7 330U/L。
Strains X1 producing phospholipase D was isolated from the soil collected from oil factory using the yolk medium plate.The activity of phospholipase D could reach to 2023 U/L in flask fermentation by TLC.Medium composition and culture condition were optimized for phospholipase D production,which was 10g/L glucose,15g/L peptone,temperature 30 ℃,pH 7.0and shaking speed of 180r/min.The maximum phospholipase D activity was increased to 7 330U/L from 2 023U/L.
