摘要
目的:检测环状RNA(circular RNA,circ RNA)在结直肠癌组织及对应癌旁组织中的表达谱变化。方法 :应用高通量circ RNA芯片技术筛选6例人结直肠癌组织及对应癌旁组织中circ RNA表达谱差异,经过对原始数据进行预处理、归一化后,筛选出差异表达的circ RNA,并对其进行聚类分析。结果:结直肠癌组织与癌旁组织之间存在892种circ RNA的差异表达,其中在肿瘤组织中高表达的有412条,低表达的有480条(变化>2倍且P<0.05)。结论:结直肠癌组织与对应癌旁组织比较,circ RNA表达谱发生了显著变化,提示这些差异表达的circ RNA可能与结直肠癌的发生发展密切相关。
Objective:To analyze the expression profile variation of circular RNA(circ RNA) between normal colorectal tissues and colorectal cancer(CRC) tissues. Methods: Human circ RNA microarray was performed to detect the difference of circ RNA expression between 6 cases of CRC tissues and 6 cases of normal colorectal tissues. After pretreatment and homogenization of raw data,the circ RNAs with differental expression were screened out and then analyzed by hierarchical clustering and scatter volcano diagram.Results: Compared with normal colorectal tissues, there were 892 circ RNAs differentially expressed which have more than 2 times variation and significant difference(P〈0.05). Among them, 412 increased and change′s fold more than 2 times,while 480 reduced and change′s fold more than 2 times. Conclusion: The circ RNA expression profile of CRC changed significantly in comparison with the normal colorectal tissues, the different circ RNAs may be associated with the development of CRC.
