摘要
以肾形亚侧耳菌株作为试验材料,利用单因素试验和正交实验方法,对其菌丝体生长发育所需要的碳源、氮源等营养条件以及pH、温度等环境条件进行研究,从而优化得出肾形亚侧耳菌丝体生长的最佳条件。结果表明:在单因素试验中,得出最理想的可利用碳源和氮源分别是果糖和蛋白胨,pH最适为6.5,温度最适为32℃。通过4因素4水平的正交实验,结果得出肾形亚侧耳的最佳培养条件为碳源为淀粉或果糖、氮源为蛋白胨、温度为28~32℃、pH为6.5。结合单因素试验和正交实验的各组菌丝长度测量结果和生长势观察结果,肾形亚侧耳的菌丝最佳培养条件为碳源为果糖、氮源为蛋白胨、温度为28~32℃、pH为6.5。
The strain of Hohenbuehelia reniformis was used as testing materials,the nutrient conditions such as carbon source and nitrogen source as well as environmental conditions such as pH and temperature required for the mycelial growth and development of this fungus were studied with the methods of single factor tests and orthogonal experiments,so that we could find the optimal conditions of mycelial growth.The results showed that,the optimum carbon source and nitrogen source were fructose and peptone,the optimum pH was 6.5 and 32℃was the optimum temperature in the single factor tests.By the orthogonal experiments with the above selected four conditions for each of the four factors,the optimal cultural condition of Hohenbuehelia reniformis was the combination of fructose and starch,peptone,with pH 6.5 and at 28℃to 32℃.In final,we get the results of the cultural condition for this stain according to the single factor tests and orthogonal experiments,the optimal cultural condition of Hohenbuehelia reniformis was the combination of fructose,peptone,with pH 6.5 and at 28℃to 32℃.
