摘要
目的探究不同制备方法对姜黄素脂质体载药量的影响,为提高姜黄素脂质体的临床应用提供依据。方法采用薄膜旋蒸法、逆向蒸发法、乙醇注入法、乙醚注入法这4种方法制备姜黄素脂质体,并利用G50凝胶过柱法纯化制备的脂质体,最后采用分光光度法测定其载药量。结果薄膜旋蒸法、逆向蒸发法、乙醇注入法、乙醚注入法的包封率依次分别为62.22%、54.78%、25.56%、28.97%,载药量分别为1.58%、1.38%、1.13%、1.36%。结论薄膜旋蒸法的包封率最高,载药量最大,为最佳制备方法,值得在姜黄素脂质体研究与应用中推广使用。
[ Objective] To investigate the effect of different preparation methods on drug loading of curcumin liposome, provide the basis for improve the clinical application of curcumin liposome.[ Methods] The curcumin liposome was treated by four preparation methods, including the thin film rotary evaporation method, the reverse phase evaporation method, the ethanol injection method and the ether injection method. G50 gel column method was applied to purify the prepared liposome, and finally the drug loading was measured by using the spectrophotometry.[ Results] The entrapment efficiency of the thin film rotary evaporation method, the reverse phase evaporation method, the ethanol injection method and the ether injection method was respectively 62.22%, 54.78%, 25.56% and 28.97%, while the drug loading was respectively 1.58%, 1.38%, 1.13% and 1.36%.[ Conclusion] Among four preparation methods, the thin film rotary evaporation method presents the highest entrapment efficiency and drug loading, which is worth promoting in research and application of curcumin liposome.
