抗CD74抗体不是汉族人群强直性脊柱炎患者的血清标志物 被引量:1

Anti-CD74 antibody is not a diagnostic marker for Chinese Han patients with ankylosing spondylitis

摘要 目的 :检测强直性脊柱炎(ankylosing spoondylitis,AS)患者血清中抗CD74抗体的水平,并评估抗CD74抗体在AS患者中的诊断价值。方法:选取确诊为AS的85例汉族患者和46名健康献血者(对照),采集血清后用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法,用2种重组人CD74蛋白和1种CD74肽作为抗原,检测抗CD74抗体水平,并采用独立样本t检验和单向方差检验进行组间均值比较。同时比较不同冻存时间的血标本中抗CD74抗体水平差异。结果:3种抗原检测出的AS患者抗CD74抗体的吸光度值(A)均值分别为0.281±0.189、0.157±0.058和0.120±0.040,与健康对照组相比,差异均无统计学意义;而AS患者中抗CD74抗体的阳性率非常低,且在不同年龄组及不同性别间抗CD74抗体水平差异无统计学意义。结论:采用不同抗原测得的抗CD74抗体水平在85例汉族AS患者中未见显著升高,考虑其在我国汉族人群中其可能并不能作为诊断AS有价值的指标。 Objective: To investigate anti-CD74 antibody level in ankylosing spondylitis(AS) patients and to evaluate the diagnostic value of anti-CD74 antibody for AS patients. Methods: Eighty-five Chinese Han patients with AS were recruited, and forty-six healthy blood donors were served as controls. Serum samples were obtained. Enzyme-linked immunosorbent assay(ELISA) was performed to detect anti-CD74 antibodies using three different antigens(two CD74 proteins and one CD74 peptide). Independent T-test and oneway Anova were used to compare the differences in anti-CD74 antibody level between AS patients and healthy controls. Results: The mean A values of three different anti-CD74 antibodies in AS patients were 0.281 ±0.189, 0.157±0.058 and 0.120±0.040, respectively, which were not significantly different from those detected in healthy controls. Positive rates of anti-CD74 antibody in AS patients were very low. The anti-CD74 antibody levels in three different age groups were not significantly different, as well as between male and female patients.Conclusions: The level of anti-CD74 antibodies are not significantly increased in Chinese Han AS patients when compared with healthy controls, and it could not be used as a diagnostic marker for Chinese AS patients.
