秦岭美容杜鹃种子组培快繁技术研究

Study on Rapid Propagation Technology of Seeds Tissue Culture of Rhododendron calophytum Franch in Qinling

以秦岭地区野生美容杜鹃种子为外植体,研究了外植体消毒时间与2种不同基本培养基初代培养,以及不同激素浓度对增殖培养和生根培养的影响。结果表明:种子消毒时间3min或者更长时间为好;在加入1.5mg/L ZT和0.5mg/L NAA的WPS培养基中发现不定芽繁殖系数达到4.0,且丛生芽长势良好;在1/2WPM中生根培养,30d生根率均达到100%,且试管苗生长健壮,根系发达。

In this study,wild Rhododendron calophytumseeds in the Qinling Mountains area were used as explants,the effect of disinfection time,two kinds of basic initial medium,hormone concentrations on the proliferation and rooting culture were explored.The results showed that the best disinfection time of seeds was 3minutes or longer.Propagation coefficient of adventitious bud reached 4.0in the WPS medium with 1.5mg/L ZT and 0.5mg/L NAA,and cluster buds grew well.Rooting culture in 1/2WPM,after 30 days,the rooting rate reached 100%.And plantlets grew strong,roots developed well.
