响应曲面法优化籽瓜干皮中L-瓜氨酸的提取工艺 被引量:3

Optimization of Extraction Process of L-citrulline from Seed Melon by Response Surface Methodology

摘要 采用超声波提取法从成熟籽瓜干皮中提取L-瓜氨酸,运用响应面法优化L-瓜氨酸提取条件。在单因素的基础上运用Box-Behnken中心组合设计以HCl体积比、料液比、提取时间3个因素3水平的试验模型,建立提取回归方程。以响应曲面法(RSM)对试验结果进行分析,对籽瓜干皮中L-瓜氨酸的提取工艺进行优化。结果表明,模拟得到的最优条件为:HCl体积比为1︰20(HCl为1 mol/L),料液比为1︰30(g/mL),提取时间70 min,此条件下L-瓜氨酸的最高提取率为2.097 99 mg/g。曲面响应法可以对籽瓜干皮中L-瓜氨酸的提取工艺进行优化。 Using the ultrasonic from the mature seed melon dry skin to extract L-citrulline, and optimize L-citrulline extraction condition by response surface method. On the basic of the single factor test, study the effects of the volume of hydrochloric acid, the ratio of material to liquid, extraction time on the L-citrulline ratio. The orthogonal test design was developed by Box-Behnken central composite test design method. Using mathematical statistical to establish relative equation, and analyze the result. Results showed that the optimized condition was as follows: the volume ratio of hydrochloric acid 1︰20(HCl 1 mol/L); the ratio of material to liquid 1︰30(g/mL), the extraction time 70 min, the highest extraction rate under this condition was 2.097 99 mg/g. The extraction technique of response surface method to seed melon dry skin L-citrulline has been developed.
