移居高原不同时间健康人群血液HIF-1α、VEGF、EPO水平的变化及意义 被引量:3

摘要 目的检测移居高原不同时间健康人血液HIF-1α、VEGF、EPO水平的情况,并探讨其意义。方法选取移居高原(平均海拔4,200m)健康汉族120例及平原(平均海拔400m)健康汉族30例。分为移居高原1周组、1月组、1年组、5年组及平原组五组,每组各30例。采用ELISA法及real-time PCR法分别检测各组血液中HIF-1α、VEGF、EPO的蛋白及mRNA表达水平,并行相关意义分析。结果移居汉族不同时间组HIF-1α、VEGF及EPO的表达均高于平原组(P<0.05),随高原低氧暴露时间的逐渐延长移居汉族HIF-1α水平逐渐下降(P<0.05),VEGF、EPO水平则随时间延长呈进行性升高(P<0.05);HIF-1α、VEGF、EPO的mRNA表达水平与其血清蛋白表达水平的变化趋势一致。结论移居高原不同时间健康人群血液VEGF及EPO水平与HIF-1α的表达变化并不完全一致。 Objective To investigate the expressions of blood hypoxia inducible factor-1α( HIF-1α),vascular endothelial growth factor( VEGF) and Erythropoietin( EPO) in healthy people who immigrating to high altitude at different time,and discusses their clinical significance.Methods 120 Han people immigrating to high altitude and 30 Han people in low altitude area were selected and divided into five groups,immigrating 1 week group,1 month group,1 year group,5 year group and low altitude group,with 30 persons in each group. HIF- 1α,VEGF and EPO protein and mRNA levels were measured using ELISA and real-time PCR methods,and analyzed the related significance.Results The levels of HIF-1α,VEGF and EPO in people who immigrating to high altitude at different time were all higher than those in low altitude group.The levels of HIF-1α in Han populations immigrating to high altitude were decreased gradually with the exposure time( P〈0.05).Levels of VEGF and EPO in immigrating Han populations were increased gradually with exposure time( P 〈0. 05). HIF- 1α,VEGF and EPO mRNA expression changed in a similar manner as their protein expressions.Conclusions The levels of blood VEGF and EPO in healthy people immigrating to high altitude at different time were not changed in a similar manner as the HIF- 1α expression.
