PDL对兔耳红色增生性瘢痕中MMP-1及TIMP-1的影响 被引量:1

摘要 目的探讨脉冲染料激光(Pulse Dye Laser,PDL)对红色增生性瘢痕组织中基质金属蛋白酶-1(Matrix Metalloproteinases-1,MMP-1)及金属蛋白酶1组织抑制剂(Tissue Inhibitor of Metalloproteinase-1,TIMP-1)表达的影响及其与增生性瘢痕消长的关系。方法将制作的84个兔耳增生性瘢痕模型分为实验组和对照组,实验组接受1次PDL治疗,对照组不接受PDL治疗;并将实验组再次分为甲组和乙组,甲组接受PDL治疗的能量密度为10 J/cm^2,乙组接受PDL治疗的能量密度为11 J/cm^2。分别于治疗前与治疗后1、2、4周切取实验组及对照组红色增生性瘢痕标本共计40个,并对其采用HE染色及免疫组织化学方法进行定性定量研究,对比研究结果。结果 PDL治疗前瘢痕组织的真皮层明显增厚,胶原纤维排列混乱;PDL治疗后瘢痕组织的真皮层变薄,胶原纤维排列稀疏,毛细血管减少。MMP-1的表达在:PDL治疗后逐渐增高,其治疗前与治疗后1、2、4周进行对比,P<0.05,差异具有统计学意义;TIMP-1的表达在PDL治疗后逐渐增高,其治疗前与治疗后1、2、4周进行对比,P》0.05,差异无统计学意义。甲组与乙组同时期MMP-1、TIMP-1的表达与对照组相比,差异均无统计学意义。结论PDL治疗后瘢痕组织中MMP-1与TIMP-1的表达均增高,且以MMP-1的表达增高更为显著,超过了TIMP-1的抑制作用,使瘢痕增生得到了有效控制。 Objective To study the effects of Pulse Dye Laser(PDL) on Matrix Metalloproteinases-1(MMP-1)and Tissue Inhibitor of Metalloproteinase-1(TIMP-1) in rabbit ear red hypertrophic scar tissues and its relationship with the growth and decline of hypertrophic scar.Methods Prepared 84 hypertrophic scar models of rabbit ears were divided into an experimental group and a control group.The experimental group was given one-time treatment of PDL while the control group wasn't given.The experimental group was further divided into Group A and Group B.The PDL energy density given in Group A was 10 J/cm^2 while that in Group B was 11 J/cm^2.Specimens of red hypertrophic scars were taken at following four time points;before the treatment,week 1/2/4 after the treatment respectively in the experimental group and the control group,40 specimens in total,on which quantitative and qualitative studies were performed by adopting HE staining and immunohistochemical methods.Results Before the PDL treatment,the dermal layer of scar tissue was remarkably thickened and the collagen fibers were arranged disorderly.After the PDL treatment,the dermis layer of scar tissue was thinner and the arrangement of collagen fibers was sparse and capillaries were decreased.The expression of MMP-1 increased gradually after the PDL treatment and the expression levels of MMP-1 on week 1,2 and 4 after the treatment were all obviously higher than that before the treatment,P〈0.05 representing the differences being statistically significant;while the expression of TIMP-1 also increased gradually after the PDL treatment but there was no statistically significant difference between the levels before the treatment and after the treatment(P〈0.05).The expression levels of MMP-1 and TIMP-1 in Group A and Group B at the same time period showed no significant difference compared with that in the control group(P〈0.05).Conclusion After the PDL treatment,the expression levels of MMP-1 and TIMP-1 in scar tissues all increased,especially the expression of MMP-1 surpassing the inhibition of TIMP-1,resulting in effective control of scar hyperplasia.
