摘要
目的分析他拉达非对肺动脉高压模型大鼠降钙素基因相关肽(CGRP)、p27及p-ERK1/2表达的影响。方法将36只雄性SD大鼠随机分为肺动脉高压(PAH)组(A组),他拉达非组(B组)及对照组(C组),各12例。观察各组大鼠血流动力学及肺血管形态学变化,以Western Blot法分析肺动脉CGRP,p27及p-ERK1/2的表达。结果与A组比,B组聚天冬氨酸(PASP)、平均肺动脉压(m PAP)显著降低(P<0.05),游离右室壁(RV)/(左室+室间隔)(LV+S)在A,B组间无显著性差异(P>0.05),但均显著高于C组(P<0.05);A组右心室肥大程度及肺血管结构变化显著高于B组(P<0.05),且均显著高于C组(P<0.05);A组大鼠CGRP,p27表达显著降低(P<0.05),p-ERK1/2表达显著上调(P<0.05),经他拉达非干预后改善,但与C组相比,差异仍有统计学意义(P<0.05)。结论他拉达非能显著改善PAH模型大鼠的血管重构,CGRP及ERK1/2/p27在其中可能扮有重要作用。
Objective To analyze the expression of CGRP,p27 and p- ERK1 / 2 in Tadalafil- intervention pulmonary arterial remodeling rats. Methods 36 male SD rats were randomly divided into PAH model group( A),Tadalafil- intervention group( B) and control group( C), the blood flow dynamics and rats pulmonary vascular morphology change were observed, the expressions of pulmonary CGRP, p27 and p- ERK1 / 2 were analyzed. Results Compared with model group, the intervention group PASP, m PAP significantly reduced( P 〈0. 05), RV /( LV + S) had no significant difference( P 〉 0. 05), but both were higher than control group( P 〈 0. 05); the model rats right ventricular hypertrophy and pulmonary vascular structural changes was significantly higher than the intervention group( P 〈 0. 05);CGRP,p27 expression in model group was significantly reduced,p- ERK1 / 2 expression significantly increased( P 〈 0. 05),after Tadalafl intervention significantly improved,but also has a significant difference with control group( P 〈 0. 05). Conclusion Tadalafil significantly improve PAH model rat vascular remodeling,CGRP,ERK1 / 2 / p27 may play an important role.
