摘要
目的 探索钙网膜蛋白在成釉细胞型颅咽管瘤的表达情况及其在肿瘤钙化形成中的作用及机制.方法 回顾性分析77例成釉细胞型颅咽管瘤患者的资料.依据肿瘤术前钙化程度将其分组.对各组标本进行钙网膜蛋白免疫组织化学检测和蛋白免疫印迹检测.同时将颅咽管瘤原代细胞进行培养后,用钙网膜蛋白siRNA转染细胞,比较转染前后培养基钙质沉积情况.结果 16例肿瘤提示无钙化,22例(29%)肿瘤轻度钙化,24例(31%)呈中度钙化,15例(20%)呈重度钙化.63例标本免疫组织化学阳性.钙网膜蛋白和钙化程度成正相关水平(r=0.853,P <0.001).蛋白免疫印迹结果和免疫组织化学检测结果保持一致.对原代细胞转染钙网膜蛋白siRNA后,培养基内钙质沉积较对照组明显下降.结论 钙网膜蛋白在钙化的成釉细胞型颅咽管瘤间质中表达,其表达和颅咽管瘤钙化程度成正相关.钙网膜蛋白在成釉细胞型颅咽管瘤钙化中机制需要更多的研究.
Objective To explore the expression of calretinin in adamantinomatous craniopharyngioma and its effect and mechanism in tumor calcification.Methods The clinical data of 77 patients with adamantinomatous craniopharyngioma were analyzed retrospectively.They were grouped according the degree of tumor calcification before tumor operation.Immunohistochemistry assay and Western blot detection of calretinin were conducted in the specimens of each group.At the same time,after the craniopharyngioma primary cells being cultured,calretinin siRNA transfected cells and the culture medium calcium deposition were compared before and after transfection.Results The tumors of 16 patients showed no calcification,22 patients had mild tumor calcification (29%),24 showed moderate calcification (31%),and 15 showed severe calcification (20%).Sixty-three specimens were positive for immunohistochemistry.Calretinin and degree of calcification showed positive correlation (r =0.853,P 〈 0.001).The results of Western blot and immunohistochemistry assay were consistent.After the primary cells being transfected into calretinin siRNA,the calcium deposition in the medium was significantly lower than that of the control group.Conclusions Calretinin expresses in the mesenchymes of calcified adamantinomatous craniopharyngioma,and its expression is positively correlated with the degree of craniopharyngioma calcification.More research is needed in the mechanisms of calretinin in the calcification of adamantinomatous craniopharyngioma.
