DHA对IFN-γ诱导的3T3L1脂肪细胞MHCⅡ及炎症分子表达的影响 被引量:1

摘要 目的探讨二十二碳六烯酸(DHA)对IFN-γ诱导的3T3L1脂肪细胞组织相容性复合体Ⅱ(MHCⅡ)及炎症分子MCP-1、IL-6、iNOS表达的影响。方法 3T3L1脂肪细胞诱导分化成成熟脂肪细胞,加入IFN-γ诱导炎症反应,再加入DHA处理,通过real time-PCR及Western blot观察DHA对分化成熟的脂肪细胞MHCⅡ及炎症分子MCP-1、IL-6、iNOS表达的影响。结果分化成熟的脂肪细胞加入IFN-γ后,MHCⅡ的相关基因Ciita及H2Ab1,炎症分子MCP-1、IL-6、iNOS明显上升,加入DHA后,Ciita、H2Ab1、iNOS、IL-6表达明显下调,MCP-1的变化比较差异无统计学意义(P>0.05)。结论 DHA能明显下调IFN-γ诱导的3T3L1成熟脂肪细胞MHCⅡ及炎症分子表达,DHA通过下调MHCⅡ基因改善脂肪细胞的炎症。 Objective To investigate the effects of DHA on expression of IFN-γinduced MHCⅡ and inflammatory cytokine in 3T3L1 adipocytes.Methods Stimulating the fully differential 3T3L1 cells with IFN-γ,we compared theexpression of MHCⅡgenes and inflammatory cytokine in the presence or absence of DHA by real-time PCR and Western blot.Results IFN-γ-mediated induction increased the expressions of Ciita,H2Ab1,iNOS,MCP-1,IL-6in 3T3L1 adipocytes.In the presence of DHA,the expressions of iNOS,IL-6and Ciita,H2Ab1 were down-regulated.Conclusion These results demonstrate that exposure to DHA reduced the level of the expressions of MHCⅡand inflammatory cytokine inmature 3T3L1 adipocytes and led to a significant reduction in inflammasome.
