摘要
选用磷脂酶D作为工具酶,在异辛烷、正丁醇和水构成的微乳体系中,进行了蛋黄磷脂酰胆碱的精制研究。结果表明,合理的工艺条件为:卵磷脂浓度为0.24 g·m L^(-1),卵磷脂与氯化胆碱质量比为7:1,加酶量为11.0 mg·m L^(-1),反应温度为40℃,反应起始pH为4.0,在该工艺条件下得到磷脂酰胆碱的转化率达84.5%。
Egg yolk lecithin was purified by phospholipase D in microemulsion system which was made by isooctane, n-butanol and water. The optimum reaction conditions were as follows: the concentration of PC was 0.24 g·mL^-l,mass ratio of PC to choline chloride was 7:1, lipase D dosage was 11.0 mg·mL^-1, reaction temperature was 40 ℃, reaction pH was 4.0. Under the optimum conditions, the conversion rate of phosphatidylcholine was 84.5%.
