茶树炭疽菌的鉴定及致病力分析 被引量:12

摘要 通过PCR扩增并测定福建省不同产地茶树上分离的炭疽病菌的β-Tub2、ITS、GDPH和ACT 4段基因序列,采用多基因系统发育分析,结合其纯培养、分生孢子、附着孢等形态特征对分离菌进行鉴定,并采用离体法测定菌株的致病力.结果表明:5株供试病原菌分离物均为Gloeosporium cingulata f.sp.camelliae,为茶树炭疽病病原菌,它们的培养性状和形态特征都相似.致病力测定表明,所有分离物均对茶树叶片致病,但其中一株致病力明显强于其余菌株,且致病力强的菌株与其余菌株的基因序列存在差异. Five isolates( tentatively named ZRG,YFS,ZHD,ABS and FHG) of anthracnose specimens from 4 counties of Fujian Province were amplified and inoculated into healthy leaves for pathogenicity test. Morphological analysis was conducted via pure cultivation,conidium andappressorium observation and subsequent β-Tub2,ITS,GPDH and ACT genes were sequenced to construct a multi-gene phylogenetic tree. Results showed that all the five pathogens,which presented similar colony and morphological features,were Gloeosporium cingulata f. sp. camelliae that causes anthracnose. All the healthy leaves were infected with anthracnose with leaf that inoculated with ZRG isolate posed the worst symptoms. This was in accordance with sequencing results that ITS of the other 4strains were significantly different from that of ZRG.
