摘要
以B16-F10小鼠黑素瘤细胞和A375人黑色素瘤细胞为模型,利用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑嗅盐法(MIT)对僵蚕(Bombyx batryticatus)内证实存在的7种化学成分(麦角甾醇、β-谷甾醇、棕榈酸、赤藓醇、甘露醇、胡萝卜苷和尿嘧啶)的抗肿瘤活性进行了初步研究.结果表明:麦角甾醇、β-谷甾醇和棕榈酸具有明显的抗肿瘤活性,并表现出明显的浓度依赖性,其中0.13 mmol/L以上处理浓度的麦角甾醇和β-谷甾醇以及0.25 mmol/L以上处理浓度的棕榈酸抑制活性最明显,高浓度的β-谷甾醇(0.5 mmol/L以上)对B16-F10的抑制活性明显高于A375.赤藓醇、甘露醇、胡萝卜苷和尿嘧啶对肿瘤细胞生长抑制作用不明显.由此初步认为僵蚕内化合物麦角甾醇、β-谷甾醇和棕榈酸在其抗肿瘤活性中具有重要作用,僵蚕可作为开发抗肿瘤药物的重要药材.
Antitumor activity of seven constituents( Ergosterol,Palmitic acid,Erythritol,Mannitol,Uracil,β- sitosterol and Daucosterol) in Bombyx batryticatus was investigated primarily by 3-( 4,5- dimethyl- 2- thiazolyl)- 2,5- diphenyl- 2- H- tetrazolium bromide( MIT); with model of tumor cell A375 and B16-F10. Results indicated that Ergosterol,β- sitosterol and Palmitic acid showed significant anti-tumor activity with a dose-dependent manner. The anti-tumor activity was most significant in case of cell treated by more than 0. 13 mmol / L( Ergosterol,β- sitosterol) and 0. 25 mmol / L( Palmitic acid).Meanwhile,Inhibition of β- sitosterol to B16-F10 was higher than that of A375 in case of high concentration( more than 0. 5 mmol / L). The growth of anti-tumor cell was not inhibited significantly when cells were in presence of different concentration of Erythritol,Mannitol,Uracil and Daucosterol,indicating no anti-tumor activity of this four components. Accordingly,Ergosterol,β- sitosterol and Palmitic acid contributed to anti-tumor activity of Bombyx batryticatus which as a traditional Chinese medicine could be used to develop into an anti-tumor medicine.
