中国北方汉族人KIR/HLA复合体基因型多态性分析

Analysis of the expression of compound KIR-HLA in Northeast Han population

摘要 目的通过分析中国北方汉族人KIR基因及配体HLA-C、HLA-Bw4基因频率的分布和识别方式,为今后进一步研究其在疾病中的作用提供理论基础。方法采用PCR-SSP和PCR-SSO技术进行KIR、HLA-C和HLA-A、-B基因分型。根据HLA-C、HLA-Bw4与KIR识别方式,分析北方汉族人HLA-Bw4配体基因型与相应激活性或抑制性KIR受体的识别情况。结果吉林汉族人群中KIR/HLA复合体的识别方式中,26.9%个体同时表达3个i KIR+HLA受配体对,51.2%个体同时表达2个i KIR+HLA,12.4%个体同时表达1个i KIR+HLA,而没有激活性受配体对的个体高达64.2%。我们还观察到83.1%的个体i KIR+HLA>a KIR+HLA,14.4%的个体i KIR+HLA=a KIR+HLA,2.5%的个体i KIR+HLA<a KIR+HLA。结论吉林汉族地区,KIR/HLA复合体基因组合中以抑制性受配体对为主,约64.2%的个体不表达激活性受配体的组合。 Objective To investigate the gene expression and molecular interaction of the human leukocyte antigen( HLA) C and killer cell immunoglobulin like receptor( KIR) genes in Jilin Han population and to provide a theoretical basis for further studies of their roles in disease. Methods A total of 201 unpaid Han blood donors were selected in Jilin. Their KIR,HLA-C and HLA-B were genotyped using PCR SSP and PCR SSO. The HLA was recognized and the corresponding stimulatory or inhibitory KIR receptor was distinguished according to the identification of HLA C1,HLA-C2,HLA-Bw4 and KIR. Results The frequencies of different numbers of KIR + HLA pairs in the Jilin Han population were: 26. 9% expressed three i KIR + HLA pairs,51. 2% expressed two i KIR + HLA pairs,and 12. 4% expressed one a KIR + HLA pair. 64. 2% did not express any i KIR + HLA pairs. Moreover,83. 1% of individuals expressed more i KIR + HLA than a KIR + HLA,14. 4%expressed simultaneously i KIR + HLA and a KIR + HLA,and 2. 5% expressed less i KIR + HLA than a KIR + HLA. Conclusion The expression of i KIR / HLA pairs is higher than the expression of a KIR / HLA pairs. In the present study,approximately 64. 2% of the individuals expressed only i KIR / HLA without a KIR / HLA in the Han population in Jilin.
