苯乙酸钠对人鼻咽癌细胞CNE-2的作用研究

摘要 运用噻唑蓝比色法(MTT法)测定苯乙酸钠对CNE-2细胞的增殖抑制作用,用流式细胞仪检测药物对细胞周期各时相的影响。评价了苯乙酸钠(NaPA)对体外培养的人低分化鼻咽癌细胞CNE-2的生长抑制作用及对细胞周期的影响。结果表明:MTT法结果显示苯乙酸钠对CNE-2细胞的增殖有一定的抑制作用;流式细胞仪检测可见用药后细胞出现凋亡,G_0-G_1期细胞增加,S期细胞减少。NaPA可以抑制CNE-2细胞的生长,呈现剂量和时间依赖性;可使细胞阻断在G_1期令S期明显减少。 The inhibitory effect and cell cycle changes of sodium phenyl acetate on nasopharyngeal carcinoma cells CNE- 2 cells in vitro were evaluated. MTT method was applied to determine the inhibition rate of cell proliferation of Na PA on CNE- 2 cells. Flow cytometry was applied to detect the influence of sodium phenylacetate on cell cycle changes of CNE- 2 cells. Results showed that the Na PA expressed various degrees of inhibition on the growth of CNE- 2 cell( P 0. 1).The IC50 was determined by MTT method. Flow cytometry displayed that the cell cycle at G_0~ G_1 phase increased when S phase decreased. And cells apoptosis was detected. The results indicated that Na PA had significantly inhibitory effect on the growth of CNE- 2 cells with dose and time dependent manner. Na PA can block CNE- 2 cells at G_1 stage.
