摘要 目的:探讨大鼠细胞色素P4502E1(CYP2E1)在非酒精性脂肪肝发生发展中的作用。方法:雄性Wistar大鼠16只随机分为2组,每组8只。对照组大鼠用普通饲料喂养,高脂组大鼠在上述方法上同时使用脂肪乳灌胃。喂养8周后处死大鼠测量体重、肝脏指数、ALT、AST、HDL-C、TG、空腹血糖(FBg)、空腹胰岛素(Ins)。大鼠肝脏组织进行HE染色,观察脂肪变及炎症坏死程度。用葡萄糖输注率观察大鼠胰岛素敏感性;用免疫组织化学染色观察CYP2E1;用逆转录聚合酶链反应(RT-PCR)测量CYP2E1的表达。结果:高脂组大鼠血清ALT、AST、TG、FBg和肝脏脂肪变程度,肝脏CYP2E1表达均高于对照组,而葡萄糖输注率低于对照组。结论:细胞色素P4502E1在大鼠非酒精性脂肪肝的发生发展中起一定的作用。 Objective:To study the role of cytochrome P4502E1(CYP2E1) in rat nonalcoholic fatty liver model. Methods: Sixteen male Wistar rats were randomly divided into control group(n=8) and high fat group(n=8). Control group was fed with normal diet, and high fat group was fed with normal diet and given a gavage of fat emulsion. All the rats were sacrificed after 8 weeks to measure the body weight, liver index, ALT, AST, HDL-C, TG, fasting glucose(Fbg) and fasting insulin(Ins). Steatosis, inflammation and necrosis in rat liver tissue were observed by HE staining. Insulin sensitivity was evaluated with the glucose infusion rate(GIR). The expression of CYP2E1 in rat liver was observed by immunohistochemical staining and measured by reverse transcriptase polymerase chain reaction(RT-PCR). Results: Several observation index in high fat group were higher than control group, which included serum ALT, AST, TG, steatosis degree, GIR and CYP2E1 expression. Conclusion:Cytochrome P4502E1 play a role in the development of rat nonalcoholic fatty live model.
