葡萄籽原花青素减轻SHR肾脏功能和结构的损伤 被引量:4

摘要 目的观察葡萄籽原花青素(GSP)对自发性高血压大鼠(SHR)肾脏功能和结构的影响及其可能机制。方法8周龄雄性SHR 24只随机分成4组,每组6只:SHR组、GSP低剂量(50 mg/kg)和高剂量治疗组(200 mg/kg)、卡托普利阳性对照治疗组(30 mg/kg)。6只同龄雄性Wistar-Kyoto大鼠设为对照组。治疗6周后,测定尾动脉收缩压(SBP);生化法检测血清尿素氮(BUN)和血肌酐(SCr)含量、肾皮质中丙二醛(MDA)含量及超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性;ELISA法检测血清胱抑素C(Cys C)含量;HE染色观察肾脏组织结构;Western blot法检测肾皮质中ERK1/2蛋白表达。结果 GSP能显著降低SHR SBP(P<0.01),改善肾功能参数、减少肾皮质中MDA含量及ERK1/2蛋白的表达(P<0.05),提高SOD和CAT活性(P<0.05),减轻肾脏结构损伤。GSP高剂量治疗组的作用尤为显著(P<0.01)。结论 GSP对SHR肾脏功能和结构损伤有缓解作用,可能与其降低SBP、抑制氧化应激和下调ERK1/2蛋白表达有关。 Objective To observe the effect of grape seed procyanidin( GSP) on renal function and structure in spontaneous hypertensive rats( SHR) and to find the possible mechanism. Methods 24 male SHR( 8-week-old) were randomized into 4 groups( n = 6 each group) : SHR,low and high dose GSP group( 50 and 200 mg / kg),captopril used for positive control group( 30 mg / kg),meanwhile,6 Wistar Kyoto rats( 8-week-old) were randomly served as control group. After 6-week treatment,tail systolic pressure,urea nitrogen( BUN) and creatinine( SCr) contents in serum,malondiadehyde( MDA) content,superoxide dismutase( SOD) and catalase( CAT) activity in the renal cortex of the rats were respectively measured. Cystain C( Cys C) level in serum was detected by ELISA.Changes of renal structure were observed after HE staining. Western blot was performed to detect the expressions of ERK1 /2 protein in the renal cortex. Results GSP significantly decreased tail systolic pressure( P〈0. 01),improve renal function parameters,reduce MDA content and the expression of ERK1 /2 in renal cortex( P〈0. 05),but increased SOD and CAT activity( P〈0. 05),relieve renal structure injury,especially in high dose GSP group( P〈0. 01). Conclusions GSP has a beneficial effect on renal function and structure injury in SHR,andits mechanism may be explained by the function of GSP in lowering SBP,inhibiting oxidative stress and reduceing expression of ERK1 /2 in renal cortex.
