白芍总苷对EAE大鼠外周免疫器官及中枢神经系统NF-κBp65表达的影响 被引量:7

Effects of total glucosides of paeony on the expression of NF-κB p65 in the peripheral immune organs and central nervous system of experimental autoimmune encephalomyelitis rats

摘要 目的观察白芍总苷(TGP)对实验性自身免疫性脑脊髓炎(EAE)大鼠外周免疫器官及中枢神经系统NF-κB p65表达的影响。方法采用后肢足掌皮内注射弗氏完全佐剂-豚鼠全脊髓匀浆,并辅以皮下注射百日咳疫苗,建立Lewis大鼠EAE模型。将大鼠随机分为对照组、模型组、TGP组,免疫后第1天起TGP组每天经口灌服白芍总苷悬浊液0.2 g/kg,对照组及模型组经口灌服等体积的生理盐水,各组第14天处死。观察各组EAE发病率、潜伏期、神经功能评分、中枢神经系统病理改变,免疫组化检测脾、淋巴结、中枢神经系统NF-κB p65的表达。结果 TGP组与模型组EAE发病率无差异(P>0.05)。与模型组相比,TGP组潜伏期延长(P<0.05),临床症状减轻(P<0.05),中枢神经系统炎症浸润细胞减少(P<0.05),脾、淋巴结、中枢神经系统NF-κB p65的表达下降(P<0.05)。结论 TGP可能通过抑制外周免疫器官及中枢神经系统NF-κB p65的表达,减轻EAE的病情。 Objective To investigate the changes of expression of NF-κB p65 protein in peripheral immune organs and central nervous system( CNS) of experimental autoimmune encephalomyelitis( EAE) rats after treated with total glucosides of paeony( TGP). Methods EAE was induced in Lewis rats by intradermal injection of guinea pig spinal cord emulsified with complete Freund’s adjuvant into hind paws,and subcutaneous injection of Bordetella Pertussis vaccine. The rats were randomly divided into control group,model group,and TGP group. From day 1 after immunization,the rats were treated with TGP suspension( oral,0. 2 g / kg) every day in TGP group and the same volume of sodium chloride in model group and control group,and all rats were executed at day 14 after immunization. The EAE incidence,latent period,neurological score and the pathological changes of CNS were determined. The protein expression of NF-κB p65 in spleen,lymph nodes and CNS was detected by immunohistochemical staining. Results There was no difference in the incidence of EAE between TGP group and model group( P 〉 0. 05). Compared with model group,the latent period was delayed in TGP group( P 〈 0. 05) and the neurological score decreased( P 〈 0. 05). Compared with model group,the number of inflammatory infiltrating cells in CNS in TGP group decreased( P 〈 0. 05). The expression of NF-κB p65 in the peripheral immune organs and CNS was lower in TGP group than in model group( P 〈 0. 05). Conclusion TGP may alleviate the state of EAE by inhibiting the expression of NF-κB p65 in the peripheral immune organs and CNS of EAE rats.
