摘要
[目的]建立一种分光光度法检测亳菊提取液总黄酮含量的方法。[方法]以亳菊为材料,采用硝酸铝络合分光光度法,在波长510 nm处间接测定总黄酮含量,比较不同提取条件下得到的总黄酮含量。[结果]当黄酮含量在0.016~0.064 g/L范围内,建立的标准曲线线性关系良好,相关系数为0.995 8,回收率试验中平均回收率为98.47%,以相对标准偏差表示的稳定性、精密度和重复性分别为1.76%、0.21%和0.25%,偏差较小。[结论]综合验证性试验发现,试验建立的定量方法简便准确,稳定性、重现性较好,数据较为合理可靠,可以作为评价亳菊提取物质量的方法。
[Objective]To establish a spectrophotometry method for detecting total flavonoids content in Chrysanthemum morifolium extract originated from Bozhou City. [Method] The content of total flavonoids was determined at 510 nm by Vis spectrophotometry using the color reaction with Al( NO3)3,the content of total flavonoids under different extraction conditions were compared. [Result]The standard curve's linear range for the UV determination is 0. 016-0. 064 g / L,with correlation coefficient being 0. 995 8 and the average recovery rate being 98. 47%. Meanwhile,the RSD representing stability,precision and repeatability are 1. 76%,0. 21% and 0. 25% which are small. [Conclusion]From the replication experiment,we can conclude this convenient and accurate method has good stability and repeatability. The data is rational and reliable.This method can be applied to assess the quality of Chrysanthemum morifolium extract.
