摘要
[目的]为海洋抗肿瘤新药的开发提供候选药物。[方法]以海洋生物虾蛄为原料,采用胰蛋白酶水解提取虾蛄软体组织中的活性多肽,采用超滤、凝胶层析、高效液相色谱等技术对其进行分离与纯化,并选择高表达肿瘤细胞株进行体外抗肿瘤活性评价。[结果]获得纯度较高的活性肽FHOS,此肽的氨基酸序列为Phe Tyr Met Asn His。采用MTT法测得FHOS对PC-3细胞和A549细胞的最高增殖抑制率达到70.1%和90.0%,呈现良好的量效和时效关系。[结论]FHOS具有一定的抑制肿瘤细胞凋亡的活性。
[Objective] The research aimed to provide candidate drugs for developing new marine anti-tumor drugs. [Method] Taking marine organism squillid as materials,active peptide was extracted from soft tissue of squillid with trypsin by enzymatic hydrolysis technique. And the hydrolysate was isolated and purified by such technologies as ultrafiltration and Sephadex G-25 gel and RP-HPLC. And its antitumor activities of every fraction obtained were detected by MTT method. [Result] A peptide( FOHS) with high purity was finally obtained. The amino acid sequence of FOHS was identified as Phe Tyr Met Asn His. The maximum proliferation inhibition rate of FHOS on PC-3 and A549 cells determined by MTT method were 70. 1% and 90. 0% respectively,showing good dose and time relationships. [Conclusion]FOHS had some activities for inhibiting the apoptosis of tumor cells.
