骨胶原肽与柠檬酸钙对去卵巢大鼠骨丢失的预防及治疗效果 被引量:7

摘要 目的将牛骨胶原肽(collagen peptides,CP)与柠檬酸钙(calcium citrate,CC)联用给去卵巢(ovariectomy,OVX)大鼠进行灌胃以评价对OVX大鼠骨丢失预防及治疗是否比单独给药更有效。方法将80只12周龄Sprague-Dawley大鼠分为预防(术后2周开始灌胃,1次/d,连续3个月)及治疗(术后3个月开始灌胃,1次/d,连续3个月)2种给药方案,每种给药方案分为以下5组(n=8):假手术组(Sham),去卵巢组(OVX),OVX大鼠服用CP组(750 mg/kg CP),OVX大鼠服用CP(750 mg/kg)和CC(75 mg/kg),去卵巢大鼠服用CC(75 mg/kg)。采用全自动生化分析仪检测血清碱性磷酸酶(alkaline phosphatase,ALP)含量,放射免疫法检测血清骨钙素(osteocalcin,OC)含量,双能X射线骨密度仪(dual energy X-ray absorptiometry,DXA)检测大鼠股骨骨密度(bone mineral density,BMD),骨生物力学测定股骨生物力学指标。结果CP-CC处理预防及治疗组OVX大鼠后呈现出骨保护效果:显著抑制OVX大鼠增加的血清ALP含量,显著抑制OVX大鼠股骨BMD的降低,显著抑制治疗组OVX大鼠股骨断裂强度的降低(P<0.05)。CP处理OVX大鼠CP后,显著抑制OVX大鼠增加的血清ALP含量,显著抑制治疗组OVX大鼠股骨BMD的降低;CC处理OVX大鼠后,显著抑制预防组OVX大鼠股骨BMD的降低,股骨断裂强度显著增加(P<0.05)。结论柠檬酸钙对去卵巢大鼠骨丢失预防具有抑制效果,而骨胶原肽联合柠檬酸钙对去卵巢大鼠骨丢失预防及治疗具有更好的效果,该结果为绝经后骨质疏松妇女骨丢失的防治提供了理论依据。 Objective To explore whether the combination of oral administration bovine collagen peptides CPs with calcium citrate( CC) is more effective than administration of either agent alone for prevention and treatment bone loss in ovariectomized( OVX) rats. Methods Eighty 12-week-old Sprague-Dawley rats were once-daily intragastric administration for 3 months after 2 weeks or 3 months of ovariectomy( OVX). The rats were randomly divided into the prevention and treatment of 2 regimens,then each regimen was divided into 5 groups( n = 8 in each group) as follows: sham+ vehicle; OVX + vehicle; OVX + 750 mg / kg CP; OVX + CP-CC( 75 mg / kg); OVX + CC( 75 mg / kg). After euthanasia,serum samples were analyzed for alkaline phosphatase( ALP) using an autoanalyzer( Model 7060; Hitachi,Tokyo,Japan) and osteocalcin( OC) using radioimmunometric assay. The femurs were analyzed for bone mineral density( BMD) by dual energy X-ray absorptiometry and bone biomechanics determination. Results OVX rats supplemented with CP-CC prevention or treatment showed osteoprotective effects: with sigfinicant reductions in the OVX-induced increases in ALP levels or decreases in femoral BMD,as well as femoral fracture strength in OVX rats with treatment( P〈0. 05). OVX rats supplemented with CP exhibited significantly inhibition in the OVX-induced increases in ALP levels or decreases in femoral BMD. OVX rats supplemented with CC exhibited significantly inhibition in the OVX-induced increases in ALP levels or decreases in femoral BMD as well as fracture strength( P〈0. 05). Conclusion Our data indicate that oral CC prevents bone loss in OVX rats and combined bovine CPs with CC prevents and treats bone loss in OVX rats. The present findings suggest that combined bovine CPs with CC is a promising alternative for reducing bone loss in osteopenic postmenopausal women.
