摘要 目的检测脆性组氨酸三联体(FHIT)和凋亡抑制蛋白Apollon(Baculoviral IAP repeat-containing protein 6,BRUCE/BIRC6)在结肠腺瘤癌变过程中mRNA的表达水平,探讨FHIT与Apollon在结肠腺瘤及结肠癌形成过程中的作用及临床意义。方法采用实时荧光定量PCR法检测30例正常结肠黏膜组织、60例结肠腺瘤组织(其中管状腺瘤48例、绒毛状腺瘤12例)、30例结肠癌组织中FHIT mRNA和Apollon mRNA的表达水平。结果 FHIT mRNA在正常对照组、结肠腺瘤组及结肠癌组中的相对表达量呈逐渐降低趋势,绒毛状腺瘤组相较于管状腺瘤组表达降低,差异均具有统计学意义(P<0.05);Apollon mRNA在正常对照组、结肠腺瘤组及结肠癌组中的相对表达量呈逐渐升高趋势,绒毛状腺瘤组相较于管状腺瘤组表达升高,差异均具有统计学意义(P<0.05);FHIT与Apollon在结肠腺瘤组和结肠癌组中的表达呈负相关(P<0.05)。结论 FHIT与Apollon在结肠癌的发生发展中发挥一定的作用,共同参与了结肠黏膜的癌变过程,联合检测两者可能有助于对结肠癌的早期诊断。 Objective The purpose of this paper is to detect the expression of FHIT and Apollon in colonic adenoma and primary colonic carcinoma,and to explore the role and clinical significance of FHIT and Apollon in the process of occurrence and development of colonic adenoma and colon cancer.Methods The reverse transcriptase real-time fluorescent quantitative PCR technique was utilized to detected the mRNA of FHIT and Apollon in a colonic adenoma group of 60 cases(tubular adenoma 48 cases and villous adenoma 12 cases),a colonic carcinoma group of 30 cases,and a normal control group of 30 cases.Results The relative expression of FHIT mRNA gradually weakened in the normal control group,the colonic adenoma group,and the colonic carcinoma group.The villous adenoma group was lower than the tubular adenoma group,with a statistically significant difference(P < 0.05); The relative expression of Apollon mRNA gradually increased in the normal control group,the colonic adenoma group,and the colonic carcinoma group.The villous adenoma group was higher than tubular adenoma,with a statistically significant difference(P < 0.05).The expression of FHIT and Apollon mRNA was negative correlated in colon carcinoma and colonic adenoma(P < 0.05).Conclusions FHIT and Apollon played a certain role in the process of occurrence and development of colonic adenoma and colon cancer,both of which participate in the process of colon mucosal canceration.The detection of FHIT and Apollon will be helpful in early diagnosis of colon cancer.
