趋化因子受体CCR4与变应性鼻炎发病机制的相关性研究 被引量:3

摘要 目的探讨CCR4与变应性鼻炎(AR)发病机制的相关性。方法用酶联免疫吸附法(ELISA)检测20例AR患者和20例健康人血清中IFN-γ、IL-4水平,流式细胞仪检测外周血CCR4+CD4+T细胞占CD4+T细胞百分比,免疫组化检测CCR4在鼻黏膜表达情况。结果与对照组比较,AR患者血清中IFN-γ的水平明显降低,差异有统计学意义(P<0.05);IL-4水平明显升高,差异有统计学意义(P<0.05);CCR4+CD4+T细胞占CD4+T细胞百分比,AR组明显升高,差异有统计学意义(P<0.05);免疫组化鼻黏膜CCR4表达水平,AR明显增高,差异具有统计学意义(P<0.05)。AR组外周血CD4+T细胞中CCR4+CD4+T细胞数量所占的百分比与血清IL-4含量呈正相关(r=0.873,P<0.05),与鼻黏膜CCR4表达量呈正相关(r=0.826,P<0.05),与血清INF-γ含量呈负相关(r=-0.801,P<0.05)。结论趋化因子受体CCR4与AR发病机制相关。 Objective To investigate the effect of CCR4 in pathogenesis of allergic rhinitis(AR). Methods The levels of IFN-γ and IL-4 in serum of 20 patients with AR and 20 healthy human were detected by ELISA, the percentage of CCR4+CD4+ T cells to CD4+ T cells in peripheral blood was detected by flow cytometry, the expressions of CCR4 in nasal mucosa were detected by immunohistochemistry. Results Compared with the control group, the levels of IFN-γ in serum of AR group were obviously decreased, the difference was statistically significant(P<0.05), the levels of IL-4 in serum increased significantly, the difference was statistically significant(P<0.05); the percentage of CCR4+CD4+ T cells to CD4+ T cells in peripheral blood of AR group were significantly increased, the difference was statistically significant(P<0.05); the expression of CCR4 in nasal mucosa of AR was significantly increased, the difference was statistically significant(P<0.05). It was positively correlated between the percentage of CCR4+CD4+ T cells to CD4+ T cells and the content of serum IL-4(r=0.873, P<0.05), as to the expression of CCR4 in nasal mucosa it was also positively correlated(r=0.826, P<0.05), while it was negative correlation with IFN-γ in serum(r=-0.801, P<0.05). Conclusion CCR4 is associated with the pathogenesis of AR.
