摘要
目的:探讨蛋白激酶D1(PKD1)对大鼠心肌梗死后心肌组织内皮型一氧化氮合酶(e NOS)及促血管生成素1(Ang1)表达调控的影响.方法:采用冠状动脉左前降支结扎造模方法复制大鼠心肌梗死模型,术后存活达2d以上的心肌梗死大鼠随机被分为模型组、PKD1处理组与CID755673处理组,每组8只.应用逆转录聚合酶链式反应(RT-PCR)分析PKD1及其阻断剂CID755673对大鼠心肌组织中e NOS及Ang1 mRNA表达的影响,应用免疫组化法和免疫印迹法分析e NOS及Ang1的蛋白表达.结果:和心肌梗死模型组相比较,PKD1处理组大鼠心肌组织中e NOS及Ang1的mRNA和蛋白表达水平显著升高(P<0.01);和PKD1处理组比较,CID755673处理组大鼠心肌组织中e NOS及Ang1的mRNA和蛋白表达水平显著下降(P<0.01),接近于模型组水平.结论:PKD1显著上调大鼠心肌梗死后受损心肌组织中e NOS及Ang1的表达.
Aim:To explore the regulation of protein kinase D1(PKD1)on the expression of endothe-lial nitric oxide synthase (eNOS)and angiopoietin-1 (Ang1)in myocardium of the rats with myocardial infarction.Methods:A myocardial infarction model of SD rats was made by ligation of the left anterior descending branch of the coronary artery.At the time of 2 days after the model of myocardial infarction was made,the rats were randomly divided into:model group,PKD1 treatment group,and CID755673 treatment group,with 8 rats in each group.Theeffects of PKD1 and CID755673 on the mRNA expres-sions of eNOS and Ang1 in myocardium of the rats with myocardial infarction were measured by reverse transcription polymerase chain reaction (RT-PCR).The protein expressions of eNOS and Ang1 were ana-lyzed by immunohistochemical and western blot.Results:Compared with those in the myocardial infarc-tion group,the mRNA and protein expressions of eNOS and Ang1 in myocardial tissue of the rats in the PKD1 treatment group were increased significantly (P 〈0.01).While compared with the PKD1 group, the CID755673group exhibited significant decrease in expressions of mRNA and protein of eNOS and Ang1 in the myocardial tissues (P 〈0.01),close to the level of the model group.Conclusion:PKD1 significantly upregulated the expression of eNOS and Ang1 in myocardium of the rats with myocardial infarction.
