摘要
[目的]探讨榛蘑多糖对三氯化铁所诱导大鼠血栓的抑制作用.[方法]将SD大鼠随机分为假手术组、模型组和用药低、高剂量组(100,400mg/kg),给予三氯化铁诱导建立大鼠颈动脉血栓模型.用药组灌胃给予不同剂量榛蘑多糖,每日1次,连续灌胃7d后检测血栓质量,ELISA法测定血浆组织纤溶酶原激活物(t-PA)、纤溶酶原激活物抑制剂(PAI-1)、6-酮-前列腺素E1α(6-keto-PGE1α)和血栓素B2(TXB2)含量,用硝酸还原酶法检测一氧化氮水平.[结果]与模型组比较,用药低、高剂量组血栓质量显著降低,t-PA、一氧化氮和6-keto-PGE1α水平均显著升高,TXB2含量显著降低(P<0.05,P<0.01).[结论]榛蘑多糖具有抗血栓作用.
OBJECTIVE To study the antithrombotic effects of Armillaria mellea polysaccharides in rat thrombosed models induced by ferric trichloride.METHODSThe carotid arterial thrombosis models were set up in SD rats,and were randomly divided to groups of control,model,low and high dose(100,400mg/kg).Rats in the treatment group were administrated by intragastric gavage with Armillaria mellea polysaccharides once per day.After 7days,the thrombus weights were measured,and the levels of t-PA,PAI-1,6-keto-PGE1αand TXB2 in plasma were ditected by ELISA,and the NO level was detected by the method of nitric acid reductase.RESULTSArmillaria mellea polysaccharides reduced significantly thrombus weight,increased significantly the levels of t-PA,NO and 6-keto-PGE1α,and reduced significantly TXB2 content as compared with the model group(P〈0.05,P〈0.01).CONCLUSION Armillaria mellea polysaccharides have an antithrombotic effects.
