桦褐孔菌多糖对脂多糖刺激小鼠巨噬细胞促炎性细胞因子表达的影响 被引量:2

摘要 [目的]观察桦褐孔菌多糖对脂多糖(LPS)刺激小鼠巨噬细胞株Raw264.7细胞与小鼠腹腔巨噬细胞促炎性细胞因子表达的影响.[方法]利用桦褐孔菌多糖(40mg/L)处理被LPS(100μg/L)刺激的Raw264.7细胞株和小鼠腹腔巨噬细胞,采用半定量RT-PCR方法测定促炎性细胞因子肿瘤坏死因子(TNF)-α和白细胞介素(IL)-1βmRNA的表达;采用ELISA法测定TNF-α和IL-1β水平.[结果]与给予LPS刺激相比较,给予桦褐孔菌多糖和LPS同时刺激的Raw264.7细胞株和小鼠腹腔巨噬细胞促炎性细胞因子TNF-α和IL-1βmRNA表达水平均明显降低(P<0.05);给予桦褐孔菌多糖和LPS同时刺激的Raw264.7细胞株在48 h后与只给予LPS刺激相比较,细胞所分泌的促炎性细胞因子TNF-α和IL-1β水平均明显下降(P<0.05).[结论]桦褐孔菌多糖对LPS刺激Raw264.7细胞株和小鼠腹腔巨噬细胞促炎性细胞因子TNF-α和IL-1βmRNA表达水平及Raw264.7细胞株促炎性细胞因子TNF-α和IL-1β的分泌均有抑制作用. OBJECTIVE To study the effect of polysaccharide from inonotus obliquus on the proinflammatory cytokine expression in the Raw 264.7cell lines and murine macrophage stimulated with lipopolysaccharide(LPS).METHODS Using the Raw 264.7cell lines and mouse peritoneal macrophage treated with polysaccharide from inonotus obliquus(40 mg/L)and stimulated with LPS(100μg/L),the expression of proinflammatory cytokines,including tumor necrosis factor(TNF)-αand interleukin(IL)-1βmRNA,was analyzed by semi-quantitative RT-PCR,and the expression of TNF-αand IL-1β mRNA was measured by ELISA.RESULTS The expression of TNF-αand IL-1βmRNA in Raw264.7 cells and mouse peritoneal macrophages stimulated simultaneously with the polysaccharide from inonotus obliquus and LPS were remarkably decreased than that stimulated with LPS(P〈0.05),and the secretion of TNF-αand IL-1βin Raw264.7 cells stimulated simultaneously with the polysaccharide from inonotus obliquus and LPS after48 h were significantly decreased than that stimulated only with LPS(P〈0.05).CONCLUSIONPolysaccharide from inonotus obliquus has inhibitory effect on the expression of TNF-α and IL-1β mRNA in Raw264.7 cells and mouse peritoneal macrophages and the secretion of TNF-αand IL-1βin Raw264.7cells by stimulated with the LPS.of can improve TNF-αand IL-1βmRNA and its protein expression induced by LPS in mouse macrophage.
