养血清脑颗粒对血管性痴呆大鼠海马CA1区BDNF和bFGF蛋白表达的影响 被引量:5

摘要 目的:观察养血清脑颗粒对血管性痴呆(VD)大鼠海马CA1区脑源性神经营养因子(BDNF)和碱性成纤维生长因子(b FGF)表达的影响。方法:72只SD大鼠随机分为假手术组、VD模型组(模型组)和养血清脑颗粒治疗组(治疗组),采用改良的Pulsinelli四血管阻断法制作VD大鼠模型,应用Morris水迷宫实验检测VD大鼠的学习记忆能力,采用免疫组化法检测海马CA1区BDNF和b FGF蛋白表达水平。结果:与假手术组比较,模型组大鼠出现明显的学习记忆障碍,各时间点海马CA1区BDNF和b FGF蛋白表达增多(P<0.01);与模型组比较,治疗组大鼠学习记忆障碍明显改善,各时间点海马CA1区BDNF和b FGF蛋白表达明显增多(P<0.01),以4周时表达最明显。结论:养血清脑颗粒能改善VD大鼠的学习记忆能力,其机制可能与养血清脑颗粒上调BDNF和b FGF蛋白表达有关。 This study was aimed to observe the effect of Yang-Xue Qing-Nao(YXQN) granules on the expression of brain-derived neurotrophic factor(BDNF) and basic fibroblast growth factor(b FGF) in CA1 area of hippocampus among vascular dementia(VD) rats. A total of 72 SD rats were randomly divided into the sham operation group, VD model group(model group) and YXQN treatment group(treatment group). The VD rat model was prepared by modified Pulsineli's four-vessel occlusion. The learning and memory abilities of rats were detected by the Morris water maze. The protein expressions of BDNF and b FGF were detected by the immunohistochemical analysis. The results showed that compared with the sham operation group, there was obvious learning and memory disorders in the model group with increased protein expressions of BDNF and b FGF in CA1 area of hippocampus at difference time points(P0.01). Compared with the model group, the learning and memory abilities of rats in the treatment group were significantly improved; and the protein expressions of BDNF and b FGF in CA1 area of hippocampus were significantly increased at difference time points(P0.01). The expression was the highest on the 4th week. It was concluded that YXQN granules can improve the learning and memory abilities of VD rats. Its mechanism may be related to the upregulation of protein expression of BDNF and b FGF.
