镇江地区儿童群体博卡病毒感染情况及基因组遗传特征

摘要 目的了解镇江地区呼吸道和胃肠道疾病患儿博卡病毒(HBoV)流行状况,并通过对代表毒株的全基因组测序了解博卡病毒遗传特征。方法收集2010年1月-2011年12月期间镇江地区156份呼吸道疾病患儿鼻咽分泌物样品和310份急性胃肠炎患儿粪便样品,采用PCR方法检测HBoV基因;选取3株镇江地区HBoV代表毒株,测序获取全基因组序列,采用CLUSTAL和MEGA4.0等软件进行序列同源性及系统发育分析。结果HBoV在鼻咽分泌物样品中的检出率为2.56%,在粪便样品检出率为3.55%;HBoV基因序列均与GenBank中提交的HBoV-1型毒株高度同源(〉95%),且与上海地区的7株HBoV-1和重庆地区的3株HBoV-1同源性最高;系统进化分析结果显示,3株HBov-1代表株聚类在三个不同的类群中,其中ZJ92与来自中国香港的HBoV毒株HK24及上海的HBoV毒株SH8遗传关系最近,ZJ42与来自泰国的毒株CU25遗传关系最近,而ZJ68与来自上海的R3080522001和SH1亲缘关系最近。结论HBoV-1为镇江地区HBoV主要流行基因型;系统进化分析显示镇江地区的HBoV-1毒株与重庆和上海的毒株有共同的分子起源。 Objective To investigate the prevalence of human bocaviruses(HBoV) in children population with gastrointestinal and respiratory diseases in Zhenjiang area,and to determine the genetic characters by the complete genome sequence of the representative strains. Methods A total of 156 nasopharyngeal aspirate fluid samplesfrom children with respiratory diseases and 310 fecal samples from children with acute gastroenteritis were collected from Hospitals in Zhenjiang from Jan. 2010 to Dec. 2011. HBoV were detected by PCR and the complete sequences of 3 representative strains were determined by sequen- cing. Phylogenetie analysis were performed using Clustal W and Mega 4.0 softwares. Results HBoV were detected in 2.56% of the nasopharyngeal secretions samples and 3.55% of the stool samples. The sequence of HBoV showed high simi- larity to HBoV-l,which shared 〉95% sequence identities with 7 strains from Shanghai and 3 strains from Chongqing. Phy- iogenetic analysis revealed that the three representative strains were clustered in three different genetic groups,in which the ZJ92 strain showed close relationship with HK24 from Hong Kong and SH8 from Shanghai, the ZJ42 showed close relation- ship with CU25 from Thailand,and the ZJ68 showed close relationship with R3080522001 and SH1 from Shanghai. Conclusion HBoV-1 was the most prevalent genotype in Zhenjiang area. Phylogenetic analysis showed the HBoV-1 strains in Zhenjiang had the common infection resources with those from Shanghai and Chongqing.
